Quantitative structure-activity relationship analysis of inhibitors of the nicotine metabolizing CYP2A6 enzyme

The purpose of this study was to develop screening and in silico modeling methods to obtain accurate information on the active center of CYP2A6, a nicotine oxidizing enzyme. The inhibitory potencies of 26 naphthalene and 16 non-naphthalene derivatives were determined for human CYP2A6 and mouse CYP2A5 enzymes. Several comparative molecular field analysis (CoMFA) models were developed to find out what types of steric and electrostatic properties are required for potent inhibitors. The IC50 values of the tested compounds varied from 0.55 to 35 400 muM for CYP2A6 and from 1 to 1500 muM for CYP2A5. The generated CoMFA models were able to accurately predict the inhibition potencies of an external test set of chemicals. Potent and specific inhibitors of the CYP2A6 enzyme can be used in the future to increase nicotine bioavailability and thus make oral nicotine administration feasible in smoking cessation therapy.

Minisci alkylation is of prime importance for its applicability in functionalizing diverse heteroarenes, which are core structures in many bioactive compounds. In alkyl radical generation processes, precious metal catalysts, high temperatures and excessive oxidants are generally involved, which lead to sustainability and safety concerns. Herein we report a new strategy using diacetyl (2,3-butanedione) as an abundant, visible light-sensitive and “traceless” hydrogen atom abstractor to achieve metal-free cross-dehydrogenative Minisci alkylation under mild conditions. Mechanistic studies supported hydrogen atom transfer (HAT) between an activated C(sp3)-H substrate and diacetyl. Moreover, with the assistance of di-tert-butyl peroxide (DTBP), the scope of the reaction could be extended to strong aliphatic C-H bonds via diacetyl-mediated energy transfer. The robustness of this strategy was demonstrated by functionalizing complex molecules such as quinine, fasudil, nicotine, menthol and alanine derivatives.

A series of pi-conjugated styryl quinolinium push-pull chromophores have been designed and synthesized in order to examine the electron-withdrawing strength of various quinolinium electron acceptor groups, and their influence on the photophysical properties and in particular on the second-order nonlinear optical response. The static molecular first hyperpolarizabilities measured by long-wavelength hyper-Rayleigh scattering are found to follow the order of the electron withdrawing strength of their acceptor groups as determined by NMR analysis. The quinolinium chromophores based on the strongest electron acceptor groups (1,2- and 1,4-dimethylquinolinium) exhibit remarkably large first hyperpolarizability values of 233 and 256 ¡Á 10-30esu respectively, which is higher than that of the well-known and widely-used pyridinium analogue 4-(4-(dimethylamino)styryl)-1-methylpyridinium 4-methylbenzenesulfonate with first hyperpolarizability = 183 ¡Á 10-30esu. The dimethylquinolinium electron acceptor groups exhibit increased electron-withdrawing strength compared to the dimethylpyridinium group used in 4-(4-(dimethylamino)styryl)-1-methylpyridinium 4-methylbenzenesulfonate, and therefore have a high potential for photonic applications.
