In a 25 mL sealed tube, (5-bromo-pyridin-3-yl)-methanol (900 mg, 4.8 mmol), 6-chloro- 3,4-dihydro-2H-isoquinolin-l-one (intermediate A-2) (800 mg, 4.4 mmol), Cul (200 mg, 1.1 mmol), CS2CO3 (3.0 g, 9.2 mmol) and (+)-(S,S)-l,2-diaminocyclohexane (0.4 mL, 3.2 mmol) were dissolved in dioxane (8.0 mL). The resulting reaction mixture was heated at 150 C for 3 hours before it was poured into H20 (50 mL) and extracted with EtOAc (2 x 125 mL). The organic layer was washed with brine, dried over anhy. Na2S04, filtered and concentrated in vacuo to give a crude product which was purified by silica gel flash chromatography (30-100% EtOAc-hexane gradient) to yield the title compound (1.1 g, 90%) as a light yellow solid. In a sealed tube, 3-bromopyridine (0.1 g, 0.633 mmol) was combined with 6-chloro-3,4- dihydroisoquinolin-l(2H)-one (intermediate A-2) (0.115 g, 633 mmol), copper (I) iodide (0.012 g, 0.063 mmol), potassium carbonate (0.175 g, 1.27 mmol) and ?,?’- dimethylethylenediamine (0.013g, 0.127 mmol) in 1,4-dioxane (2 mL). The reaction mixture was heated at 110 C over night. The mixture was cooled to room temperature, filtered through dicalite and washed with DCM. The residue was purified by silica gel flash chromatography eluting with a 5 to 100% EtO Ac-heptane gradient to give the title compound (0.107 g, 65 %) as a white solid.
