On the basis of structure-activity relationship (SAR) studies, as well as the stability and availability of the starting materials of those compounds, (S)-N-[1-(2-fluorophenyl)-3,4,6,7-tetrahydro-4-oxo-pyrrolo[3,2,1-jk][1 ,4]benzodiazepin-3-yl]1 H-indole-2-carboxamide (9f, FK-480) was selected as a candidate compound for further evaluation. The absolute configuration of the precursor of FK-480, (3S)-amino-1,4-benzodiazepine derivative ((S)-8a, R1 = F) was determined by an X-ray crystallographic study of its ureido derivative with (S)-alpha-methylbenzyl isocyanate. A simple and practical copper catalyzed C-4 carboxylation reaction of 1-naphthylamide derivatives using carbon tetra bromide and methanol is reported here. Picolinamide and its derivatives are used as a bidentate directing group for the distal C4-H functionalization. Various substituted naphthylamide derivatives, and anilides are employed for the carboxylation and proceeds in good yields under mild condition. From the outcome of experimental results, it is proposed that C4-H carboxylation reaction of 1-naphthylamides might proceed through a single electron transfer (SET) pathway. (Figure presented.).

To elucidate the structural, spectroscopic and bonding properties, the theoretical calculations at the DFT level were undertaken for 1, 3, 5 and 7. The synthesized complexes exhibited moderate activity in the oxidation of 1-phenylethanol and certain alkanes (n-heptane and methylcyclohexane) with tert-butyl hydroperoxide (TBHP) in acetonitrile. Chromatograms of products obtained from the alkanes indicated that a sufficient sterical hindrance exists around of the rhenium catalytic center.

Anti-inflammatory and anti-migratory activities of isoquinoline-1-carboxamide derivatives in lps-treated bv2 microglial cells via inhibition of MAPKS/NF-KappaB pathway

Eleven novel isoquinoline-1-carboxamides (HSR1101~1111) were synthesized and evaluated for their effects on lipopolysaccharide (LPS)-induced production of pro-inflammatory mediators and cell migration in BV2 microglial cells. Three compounds (HSR1101~1103) exhibited the most potent suppression of LPS-induced pro-inflammatory mediators, including interleukin (IL)-6, tumor necrosis factor-alpha, and nitric oxide (NO), without significant cytotoxicity. Among them, only N-(2-hydroxyphenyl) isoquinoline-1-carboxamide (HSR1101) was found to reverse LPS-suppressed anti-inflammatory cytokine IL-10, so it was selected for further characterization. HSR1101 attenuated LPS-induced expression of inducible NO synthase and cyclooxygenase-2. Particularly, HSR1101 abated LPS-induced nuclear translocation of NF-kappaB through inhibition of IkappaB phosphorylation. Furthermore, HSR1101 inhibited LPS-induced cell migration and phosphorylation of mitogen-activated protein kinases (MAPKs) including extracellular signal-regulated kinase 1/2, c-Jun N-terminal kinase, and p38 MAPK. The specific MAPK inhibitors, U0126, SP600125, and SB203580, suppressed LPS-stimulated pro-inflammatory mediators, cell migration, and NF-kappaB nuclear translocation, indicating that MAPKs may be the upstream kinase of NF-kappaB signaling. Collectively, these results demonstrate that HSR1101 is a potent and promising compound suppressing LPS-induced inflammation and cell migration in BV2 microglial cells, and that inhibition of the MAPKs/NF-kappaB pathway mediates its anti-inflammatory and anti-migratory effects. Based on our findings, HSR1101 may have beneficial impacts on various neurodegenerative disorders associated with neuroinflammation and microglial activation.

An 8,5-fused bicyclic peptidomimetic ring system generated by a stereoselective ring metathesis reaction was elaborated into potent inhibitors of interleukin-1beta converting enzyme (ICE, caspase-1). Multiple compounds were found that exhibited ICE IC50 values <10 nM and were selective over caspase-3 and caspase-8. These active analogs generally possessed good activity (IC50 values <100 nM) in a whole cell assay measuring IL-1beta production. Synthesis and Crystal Structures of [(Nicotinic Acid)2H] +I-, [(2-Amino-6-methylpyridine)H]+(NO 3)-, and the 1:1 Complex Between 1-Isoquinoline Carboxylic Acid (Zwitter Ion) and L-Ascorbic Acid Assembled via Hydrogen Bonds

Three new complexes, namely [(nicotinic acid)2H] +I-, [(2-amino-6-methylpyridine)H]+ (NO 3)-, and the 1:1 complex between 1-isoquinoline carboxylic acid (zwitter ion form) and L-ascorbic acid were synthesized. The IR spectra revealed different types of hydrogen bonds in these compounds. The X-ray structure determination has shown the first compound to consist of a packing of [(nicotinic acid)2H]+ cations and I – anions. In the dimeric cation the two nicotinic acid molecules (zwitter ions) are connected through hydrogen bonds (O-H…O). Each dimer is further engaged in other hydrogen bonds with adjacent dimers giving 2D layers. The I- ion is located at the inversion center. In the second compound the cation and anion are connected via hydrogen bonds formed between oxygen atoms of the NO3- anion and NH and NH2 of the cation generating a layer structure. All atoms are coplanar on mirror planes. In the 1:1 complex the two molecules are connected through hydrogen bonds formed between the two oxygen atoms of the carboxylate group of 1-isoquinoline carboxylic acid (zwitter ion) and the oxygen atoms of the two adjacent hydrogen groups of the L-ascorbic acid molecule. These complex molecules are engaged in other hydrogen bonds with each other forming a 2D system normal to the long b-axis of the unit cell.

Aluminum-catalyzed asymmetric alkylations of pyridyl-substituted alkynyl ketones with dialkylzinc reagents

Alkylations of pyridyl-substituted ynones with Et2Zn and Me 2Zn, promoted by amino acid-based chiral ligands in the presence of Al-based alkoxides, afford tertiary propargyl alcohols efficiently in 57% to >98% ee. Two easily accessible chiral ligands are identified as optimal for reactions of the two dialkylzinc reagents. Catalytic alkylations with Et 2Zn require a chiral ligand carrying two amino acid moieties (valine and phenylalanine) along with a p-trifluoromethylphenylamide C-terminus. In contrast, reactions with Me2Zn are most effectively promoted in the presence of a chiral ligand containing a single amino acid (benzyl cysteine), capped by an n-butylamide. Enantiomerically enriched tertiary alcohols bearing a pyridyl and an alkyne substituent can be functionalized in a variety of manners to furnish a wide range of difficult-to-access acyclic and heterocyclic structures; two noteworthy examples are Cu-catalyzed protocols for conversion of tertiary propargyl alcohols to enantiomerically enriched tetrasubstituted allenes and bicyclic amides that bear an N-substituted quaternary carbon stereogenic center. Mechanistic models that account for the trends and enantioselectivity levels are provided.
