The complexation of VIVO2+ ion with 10 picolinate and quinolinate derivatives, provided with the donor set (N, COO-), was studied in aqueous solution and in the solid state through the combined application of potentiometric (pH-titrations), spectroscopic (EPR, UV/vis and IR spectroscopy), and computational (density functional theory (DFT) calculations) methods. Such derivatives, that form potent insulin-enhancing VIVO2+ compounds, are picolinic (picH), 6-methylpicolinic (6-mepicH), 3-methylpicolinic (3-mepicH), 5-butylpicolinic or fusaric (fusarH), 6-methyl-2,3-pyr-idindicarboxylic (6-me-2,3-pdcH2), 2-pyridylacetic (2-pyacH), 2-quinolinecarboxylic or quinaldic (quinH), 4-hydro-xyquinoline-2-carboxylic or kynurenic (kynurH), 1-isoquinolinecarboxylic (1-iqcH) and 3-isoquinolinecarboxylic (3-iqcH) acid. On the basis of the potentiometric, spectroscopic, and DFT results, they were divided into the classes A, B, and C. The ligands belonging to class A (3-mepicH, 1-iqcH, 2-pyacH) form square pyramidal complexes in aqueous solution and in the solid state, and those belonging to class B (picH, fusarH, 3-iqcH) form cis-octahedral species, in which the two ligands adopt an (equatorial-equatorial) and an (equatorial-axial) arrangement and one water molecule occupies an equatorial site in cis position with respect to the V=O bond. Class C ligands (6-mepicH, 6-me-2,3-pdcH2, quinH, kynurH) yield bis chelated species, that in water are in equilibrium between the square pyramidal and frans octahedral form, where both the ligand molecules adopt an (equatorial-equatorial) arrangement and one water is in trans position with respect to the V=O group. The frans-octahedral compounds are characterized by an anomalous electron paramagnetic resonance (EPR) response, with Az value being reduced by about 10% with respect to the prediction of the “additivity rule”. dFt methods allow to calculate the structure, 51V hyperfine coupling constant (Az), the stretching frequency of V=O bond (vV=O), the relative stability in aqueous solution, and the electronic structure and molecular orbital composition of bis chelated complexes. The results were used to explain the biotransformation of these potent insulin-enhancing compounds in blood serum.

Comparative equilibrium and structural studies of new pentamethylcyclopentadienyl rhodium complexes bearing (O,N) donor bidentate ligands

Complex formation processes of the (O,N) donor ligands 6-methylpicolinic acid (6-Mepic), quinoline-2-carboxylic acid (2-QA) and 3-isoquinolinecarboxylic acid (3-iQA) with the organometallic moiety (eta5-pentamethylcyclopentadienyl)rhodium(III) (RhCp*) were studied in aqueous solution by the combined use of pH-potentiometry, 1H NMR spectroscopy and UV-Vis spectrophotometry. The solid phase structures of the [RhCp*(L)Cl] complexes bearing 6-Mepic and 2-QA were characterized by single-crystal X-ray diffraction analysis. Studies revealed the exclusive formation of mono complexes of the form [RhCp*(L)(H2O)]+ (L = deprotonated form of the ligands) and [RhCp*(L)(OH)]. The positively charged aqua species predominate at physiological pH even in the micromolar concentration range. The H2O/Cl? co-ligand exchange constants showed that all complexes preferably retain the chlorido ligand at the third coordination site at chloride ion concentrations present in the serum. In addition in vitro cytotoxicity of these [RhCp*(L)Cl] complexes was evaluated in three human cancer cell lines (A549, SW480 and CH1/PA-1) where they showed minor cytotoxic potency.

New trisubstituted 1,2,4-triazoles as ghrelin receptor antagonists

Ghrelin receptor ligands based on a trisubstituted 1,2,4-triazole scaffold were recently synthesized and evaluated for their in vitro affinity for the GHS-R1a receptor and their biological activity. In this study, replacement of the alpha-aminoisobutyryl (Aib) moiety (a common feature present in numerous growth hormone secretagogues described in the literature) by aromatic and heteroaromatic groups was explored. We found potent antagonists incorporating the picolinic moiety in place of the Aib moiety. In an attempt to increase affinity and activity of our lead compound 2, we explored the modulation of the pyridine ring. Herein we report the design and the structure-activity relationships study of these new ghrelin receptor ligands.
