Light-Driven Intramolecular C?N Cross-Coupling via a Long-Lived Photoactive Photoisomer Complex

Reported herein is a visible-light-driven intramolecular C?N cross-coupling reaction under mild reaction conditions (metal- and photocatalyst-free, at room temperature) via a long-lived photoactive photoisomer complex. This strategy was used to rapidly prepare the N-substituted polycyclic quinazolinone derivatives with a broad substrate scope (>50 examples) and further exploited to synthesize the natural products tryptanthrin, rutaecarpine, and their analogues. The success of gram-scale synthesis and solar-driven transformation, as well as promising tumor-suppressing biological activity, proves the potential of this strategy for practical applications. Mechanistic investigations, including control experiments, DFT calculations, UV-vis spectroscopy, EPR, and X-ray single-crystal structure of the key intermediate, provides insight into the mechanism.

Synthesis of 1-(pyridyl, quinolyl, and isoquinolyl)azulenes by Reissert-Henze type reaction

Azulene derivatives reacted with N-oxide of several heterocycles in the presence of trifluoromethanesulfonic anhydride (Tf2O) to afford 1-(pyridyl, quinolyl, and isoquinolyl)azulenes in good yield, respectively. In the case of the reaction with the 1-azulenyl methyl sulfide (12), 1,1?-biazulene derivative 13 was obtained under the similar reaction conditions. The first synthesis of unsymmetrical 1,3-di(pyridyl)azulene derivative was also established via our new preparation method following the electrophilic pyridinylation using the reaction with pyridine in the presence of Tf2O.

Bacterial resistance is increasing rapidly, requiring urgent identification of new antibacterial drugs that are effective against multidrug-resistant pathogens. Novel bacterial topoisomerase inhibitors (NBTIs) provide a new strategy for investigating the well-validated DNA gyrase and topoisomerase IV targets while preventing cross-resistance issues. On this basis, starting from a virtual screening campaign and subsequent structure-based hit optimization guided by X-ray studies, a novel class of piperazine-like NBTIs with outstanding enzymatic activity against Staphylococcus aureus and Escherichia coli DNA gyrase and topoisomerase IV was identified. Notably, compounds (¡À)-33, (¡À)-35, and (¡À)-36 with potent and balanced multitarget enzymatic profiles exhibited excellent efficacy against selected Gram-positive and Gram-negative pathogens, as well as clinically relevant resistant strains. Overall, the new NBTI chemotype described herein, owing to the broad-spectrum antibacterial activity and favorable in vitro safety profile, might serve as a basis for the development of novel treatments against serious infections.

4-Methyl- and 4-benzyl-5-aminoisoquinolin-1-ones are close analogues of the water-soluble PARP-1 inhibitor 5-AIQ. N-Benzhydryl-N-cinnamyl-2-iodo-3-nitrobenzamide similarly gave 2-benzhydryl-4-benzyl-5-nitroisoquinolin-1-one and 2-benzhydryl-4-benzylidene-5-nitro-3,4-dihydroisoquinolin-1-one. The isomeric products are not interconvertible. A deuterium-labelling study indicated that the isomers were formed by different pathways: a pie-allyl-Pd route and the classical Heck route. The synthesis and optimization of the anti-Helicobacter pylori activity of a novel series of benzyloxyisoquinoline derivatives that was discovered by a random screening process, are described. In the in vitro assay, compound 10c containing a 3-acetamido-2,6-dichlorobenzyl substituent was found to have extremely potent activity against H. pylori and no activity against other common bacteria. The anti-H. pylori activity of 10c was superior to that of amoxicillin (AMPC) (1) and clarithromycin (CAM) (2). A novel high-efficiency white hyperbranched polymer derived from polyfluorene with green and red iridium(III) complexes as the cores

A new kind of single triple-color hyperbranched polymer PF-Ir(ppy)2(pytzph)m-Ir(piq)2(pytzph)n was designed and synthesized, in which green and red iridium(III) complexes acted as cores and blue polyfluorene acted as backbone. PF-Ir(ppy)2(pytzph)m-Ir(piq)2(pytzph)n (m = 5, n = 5 and m = 25, n = 5) had higher quantum efficiency than PF-Ir(piq)2(pytzph)5. The energy transfer from green core bis(2-phenylpyridine)[3-(2-pyridyl)-5-phenyl-1,2,4-triazole]iridium(III) [Ir(ppy)2(pytzph)] with high triplet energy level (ET = 2.58 eV) to red core bis(1-phenylisoquinolinato)[3-(2-pyridyl)-5-phenyl-1,2,4-triazole]iridium(III) [Ir(piq)2(pytzph)] (ET = 2.12 eV) could realize by intermediate poly(9,9-dioctylfluorene) (PF) segments. A typical single emitting-layer device with the configuration of ITO/PEDOT:PSS (40 nm)/PF-Ir(ppy)2(pytzph)m-Ir(piq)2(pytzph)n (80 nm)/TPBi (40 nm)/LiF (0.8 nm)/Al (100 nm) was fabricated. Among the white polymers, PF-Ir(ppy)2(pytzph)5-Ir(piq)2(pytzph)5 exhibited excellent electroluminescent properties with a Commission Internationale Ed I’eclairage (CIE) coordinate of (0.32, 0.34) and a maximum luminous efficiency of 11.49 cd/A.

Evaluation of the O- methyl ether derivative of 4 suggested that the 3-hydroxymethyl substituent might be involved in a hydrogen-bond donor-type of interaction at a sterically compact region in the PNMT active site. The directionality of the steric bulk tolerance at both the PNMT active site and the alpha2-adrenoceptor appears to be the same. Since the presence of a hydrophilic electron- withdrawing substituent (such as NO2, SO2CH3, or SO2NH2) at the 7- position of THIQ reduced the binding affinity toward the alpha2-adrenoceptor, we investigated the combination of both a hydrophilic electron-withdrawing 7- substituent and a 3-alkyl substituent on a THIQ nucleus. A synergistic effect in increasing the PNMT-inhibitory potency of the THIQ nucleus and reducing the affinity toward the alpha2-adrenoceptor was observed with this 3,7- disubstitution. Remarkably, 7-aminosulfonyl-3-hydroxymethyl-THIQ (12; PNMT K(i) = 0.34 muM, alpha2 K(i) = 1400 muM, selectivity = 4100) displayed a 23- 680-fold enhanced selectivity over the parent compounds 27 (SK and F 29661; PNMT K(i) = 0.55 muM, alpha2 K(i) = 100 muM, selectivity = 180) and 4 selectivity = 6.0) and is thus the most selective PNMT inhibitor yet reported.

Iridium complexes containing three different ligands as white OLED dopants

Previously, we studied the luminescence characteristics of Ir(ppy)2(F2-ppy) and Ir(ppy)2(piq-F), which are heteroleptic iridium complexes involving two kinds of ligands, where F2-ppy, ppy and piq-F represent 2-(2,4-difluoro-phenyl)- pyridine, 2-phenylpyridine and 1-(4′-fluoroyphenyl)isoquinoline, respectively. Photoluminescence (PL) spectrum of Ir(ppy)2(F2-ppy) showed an emission peak at 495nm in bluish green area. Ir(ppy)2(piq-F) showed two peaks at 513nm and 600nm in the PL spectrum. In order to make a white phosphorescent emitter for OLED, we herein designed a heteroleptic iridium complex containing three different ligands. We thought that reaction of F2-ppy, ppy and piq-F ligands all together with Ir(acac)3 might lead to Ir(F2-ppy)(ppy)(piq-F) among the mixture, displaying red and bluish green emission simultaneously. Ir(F2-ppy)(ppy)(piq-F) and other double-heteroleptic complex mixture are prepared from various concentration combination of ligands. The heteroleptic iridium complex mixture displays a variety of emission color, depending upon the combination ratio of ligands in the iridium complex synthesis. UV-vis absorption and photoluminescence (PL) spectra of the mixtures are analyzed and compared with the double-heteroleptic complexes. The white-yellow emission is observed when the mole ratio of ligands is 5:1:3 for F2-ppy, ppy and piq-F, respectively.
