A Borrowing Hydrogen Strategy for Dehydrative Coupling of Aminoisoquinolines with Benzyl Alcohols in Water

We report a borrowing hydrogen strategy for a palladium-catalyzed dehydrative coupling of aminoisoquinolines with benzylic alcohols in water. This cascade reaction using the pi-benzylpalladium system can be achieved in an atom-economic process without the need for base or other additives, furnishing the N-benzylated aminoisoquinolines in moderate to excellent yields along with water as the sole co-product. The crossover experiment using [D7]benzyl alcohol and 4-methoxybenzyl alcohol afforded H/D scrambled products. KIE experiments showed that benzylic C?H bond cleavage of benzyl alcohol was involved in the turnover limiting step (KIE = 4.4). The coupling reaction was found to be first order in benzyl alcohol with a kinetic solvent isotope effect (KSIE) of 1.6. These experimental results are consistent with a borrowing hydrogen mechanism in water. Notably, the water-soluble Pd0/TPPMS system can be applied to the more challenging catalytic benzylic amination with aminoisoquinoline nucleophiles despite the possible deactivation of PdII species.

The oxidation of 2-substituted pyridines and selected N-containing aromatic heterocycles by dimethyldioxirane (1) produced the corresponding N-oxides as the sole products, quantitatively in most cases. The second order rate constants for N-oxidation by 1 in dried acetone at 23C were determined for a series of 2-substituted pyridines 2-10, quinolines 11-14 and isoquinolines 15,16. An excellent correlation of log k2 with Taft (sigma*) constants was obtained for 2-substituted pyridines (R = Me, Et, Prn, Pr i, 3-pentyl) with the exception of the data for 2-f-butylpyridine. The results for the substituted quinolines and isoquinolines followed the same trends observed with the pyridines. Steric effects due to 2-substitution and periinteractions can substantially reduce reactivity. The results provide insights into the geometrical requirements for N-oxidation by dimethyldioxirane.

The highly regioselective electrophotocatalytic C-H functionalization of ethers is described. These reactions are catalyzed by a trisaminocyclopropenium (TAC) ion at mild electrochemical potential with visible light irradiation. Ethers undergo oxidant-free coupling with isoquinolines, alkenes, alkynes, pyrazoles, and purines with typically high regioselectivity for the less-hindered alpha-position. The reaction is proposed to operate via hydrogen atom transfer (HAT) from the substrate to the photoexcited TAC radical dication, thus demonstrating a new reactivity mode for this electrophotocatalyst.
