The enantiomerically pure tetrahydroisoquinolines (S)- (-)-norcryptostyline I [(S)-(-)-1], (S)-(-)-norcryptostyline II [(S)-(-)-2], (R)-(+)-salsolidine [(R)-(+)-3] and (S)-(-)-norlaudanosine [(S)-(-)-4] were then obtained in 45%, 40%, 41% and 38% yields, respectively, via resolution of the racemic compounds (RS)-(±)-1-4 with half equivalent of chiral acids. In addition, the enantiomerically enriched compounds (R)-(+)-1, (R)-(+)-2, (S)-(-)-3 and (R)-(+)-4 from the mother liquors were efficiently racemized via a one-pot redox method in almost quantitative yields.

The oxidation of water by [Ru(DIQ-Bpy)(H2O)]2+ was observed in the presence of excess amounts of CeIV. Relative to [Ru(DPA-Bpy)(H2O)]2+ [DPA-Bpy = N,N-bis(2-pyridinylmethyl) -2,2-bipyridine-6-methanamine], the substitution of pyridine groups in DPA-Bpy with electron-withdrawing isoquinolines results in higher redox potential and lower activity for the oxidation of water by [Ru(DIQ-Bpy)(H2O)] 2+. A kinetic study of water oxidation by [Ru(DPA-Bpy)(H 2O)]2+ suggests a mononuclear pathway for the oxidation of water. The noncovalent interaction between isoquinoline groups in [Ru(DIQ-Bpy)(H2O)]2+, which favors the formation of dinuclear species, might account for the lower activity for water oxidation by [Ru(DIQ-Bpy)(H2O)]2+. Mononuclear Ru complexes with a pentadentate ligand, N,N-bis[(isoquinolin-1-yl)methyl][6-(pyridin-2-yl)pyridin- 2-yl]methanamine (DIQ-Bpy), were synthesized and characterized. From this lead, the key pharmacophore groups for delta receptor affinity and activation were more clearly defined by SAR and mutagenesis studies. Further structural modifications on the basis of 1 confirmed the importance of the N,N-diethylbenzamide group and the piperazine lower basic nitrogen for delta binding, in agreement with mutagenesis data. A number of piperazine N-alkyl substituents were tolerated. In contrast, modifications of the phenyl group led to the discovery of a series of diarylmethylpiperazines exemplified by N,N-diethyl-4-[1-piperazinyl(8-quinolinyl)-methyl]benzamide (56) which had an improved in vitro binding profile (IC50 = 0.5 nM, mu/delta = 1239, EC50 = 3.6 nM) and increased in vitro metabolic stability compared to SNC-80.

A bulky alkyl substituent in the C-8 position of the tetrahydroisoquinoline produces a clear increase in the affinity for the apamin sensitive binding sites. The presence of an electron-withdrawing group in the C-5 and C-8 positions is not a suitable substitution for the affinity of drugs structurally related to N-methyl-laudanosine. Thiophenic analogues and 8-methoxy derivatives possess a poor affinity for the apamin sensitive binding sites. Electrophysiological studies performed with the most effective compound showed a blockade of the apamin sensitive afterhyperpolarization in rat dopaminergic neurons.
