Synthesis and photophysical insights of new fused N-heterocyclic derivatives with isoquinoline skeleton

Six new fused isoquinoline based compounds (compounds 5a-c with pyrrolo[2,1-a] isoquinoline structure and compounds 6a-c with imidazo[2,1-a]isoquinoline skeleton) have been synthesized using the [3 + 2] cycloaddition of the several in situ generated cycloimmonium ylides to ethyl propiolate or ethyl cyanoformate. All the synthesized compounds have been investigated in solution by UV-VIS absorption, steady and time-resolved fluorescence methods. The effect of the substituents on the spectral characteristics has been demonstrated. These derivatives displayed an intense emission between 360 and 420 nm. The emission quantum yields (Phi = 0.54-0.64) of pyrroloisoquinoline derivatives in dimethylsulfoxide (DMSO) were significantly higher than those of imidazoquinolines (0.03-0.16). The fluorescence decay of isoquinoline derivatives follows a biexponential law. A noticeable response of these isoquinoline derivatives to sodium hydroxide was observed. (C) 2020 Elsevier B.V. Selective removal of isoquinoline and quinoline from simulated fuel using 1,1 ‘-binaphthyl-2,2 ‘-diol (BINOL): crystal structure and evaluation of the adduct electronic properties

1,1′-Binaphthyl-2,2′-diol/quinoline (BINOL/QUN) and 1,1′-binaphthyl-2,2′-diol/isoquinoline (BINOL/ISOQUN) adducts were successfully synthesized. X-ray single crystals of BINOL/QUN and BINOL/ISOQUN were grown and analysed. The crystal packing of the molecules in both adducts confirmed that they are held in aggregates by strong hydrogen bonds (O2-H2 center dot center dot center dot O3), (O3-H3 center dot center dot center dot N1), (O2-H2 center dot center dot center dot O1), (O1-H1 center dot center dot center dot N1), (O2-H2 center dot center dot center dot O1) and weak hydrogen C-H center dot center dot center dot pi bonds. The patterns of the hydrogen bonding network as well as the conformation of BINOL contribute to the formation of the shape of the voids that entrap quinoline and isoquinoline. Molecular modelling which was employed to investigate the electronic properties of BINOL/QUN and BINOL/ISOQUN shows that the HOMO positions of the adducts are localized around the 1,1′-binaphthyl-2,2′-diol (BINOL), while the LUMO is positioned on isoquinoline and quinoline. Thermodynamic parameters obtained from isothermal titration calorimetry (ITC) revealed a stronger isoquinoline/BINOL interaction compared to quinoline/BINOL. 6-Vinyl-1,1′-binaphthyl-2,2’-diol was co-polymerized with styrene to form [DBN-co-STY]. Electrospun [DBN-co-STY] exhibited selectivity for quinoline and isoquinoline in a model simulated fuel presenting an adsorption capacity of 2.2 and 2.4 mg g(-1) respectively. The adsorption study showed a higher adsorption capacity for isoquinoline compared to quinoline. This may be attributed to the more favourable electronic properties (HOMO-LUMO properties) of isoquinoline. This concept demonstrates the possibility of extracting/separating isoquinoline and quinoline from fuel.

The effect of isoquinoline alkaloids on opiate withdrawal

Our interest has been centered on isoquinoline alkaloids obtained from Argemone mexicana (Papaveraceae), Aristolochia constricta (Aristolochiaceac) and the opium alkaloid, papaverine. In this respect, the effect of these isoquinoline alkaloids was investigated on contractions induced by naloxone of isolated guinea pig ileum acutely exposed to morphine in vitro. The activity of these alkaloids was compared to the control compound, papaverine. Furthermore, the effect of these isoquinoline alkaloids was also determined on naloxone-precipitated withdrawal in isolated guinea pig ileum exposed to DAMGO (highly selective mu opioid receptor agonist) and U50-488H (highly selective kappa opioid receptor agonist) to test whether the possible interaction of isoquinoline alkaloids on opioid withdrawal involves mu-and/or kappa-opioid receptors. Isoquinoline alkaloids from A. mexicana (from 5x 10(-6) to 1 x 10(-4) M), from A. constricta (1x 10(-5) -5x 10(-5) -1x 10(-4) M) as well as papaverine treatment (1×10(-7) -5×10(-6) -1×10(-6) M) before or after the opioid agonists were able of both preventing and reversing the naloxone-induced contraction after exposure to mu(morphine and DAMGO) or kappa (U50-488H) opiate receptor agonists in a concentration-deperident manner. Both acetylcholine response and electrical stimulation were also reduced by isoquinotine alkaloids and papaverine treatment as well as the final opiate withdrawal was still reduced. The results of the present study indicate that isoquinoline alkaloids as well as papaverine were able to produce significant influence on the opiate withdrawal in vitro and these compounds were able to exert their effects both at muand kappa opioid agonists.

Inhibitory effect of isoquinoline alkaloids on movement of second-stage larvae of Toxocara canis

To find new anthelmintics against parasites living in host tissues, we used an in vitro assay to screen isoquinoline alkaloids for nematocidal activity on the larva of dog roundworm, Toxocara canis. To evaluate the efficacy of anthelmintics in vitro, Tsuda et al. previously introduced the concept of Relative Mobility (RM) of Toxocara larvae. After improvement of the assay system using image data processing, we generated a new index, RM50, the concentration at which RM=50%. However, except for pyrantel, the RM, of most existing anthelmintics could not be calculated because of low activity. Of the isoquinoline alkaloids tested, emetine, sanguinarine, 6-methoxydihydrosanguinarine (6-MS), chelerythrine and berberine showed strong nematocidal activities. However, these compounds were highly cytotoxic; thus, the prospect of their direct application is low. We then tested the cytotoxicity (IC50) of other isoquinoline alkaloids in HL60 tissue-culture cells. We continued our search for new anthelmintics by examining in detail the relationship between RM50 and IC50. We determined that an ideal target compound would exhibit a low RM50/IC50 ratio. Allocryptopine, dehydrocorydaline and papaverine were identified as potentially effective anthelmintics.
