Iodinated contrast media (ICMs) are a class of X-ray contrast media worldwide utilized for radiog. procedures. Since they cannot be removed efficiently during water treatment, they can be found in surface and groundwater. In this work, a rapid and sensitive direct injection liquid chromatog.-tandem (LC-MS/MS) method for the simultaneous anal. of seven ICMs media (iopamidol, ioxitalamic acid, diatrizoic acid, iothalamic acid, iohexol, iomeprol and iopromide) in complex aqueous matrixes has been developed and validated. The MDLs for the analytes ranged from 0.7 to 21 ng L-1 in ultrapure water, and recoveries ranged from 86 to 100% in drinking water, 85-103% in groundwater and 84-105% in WWTP effluent. A stereo-isomer for iopromide was separated This analytic method was applied to investigate the removal of target ICMs by low pressure ultra violet light (LPUV) advanced oxidation processes with three oxidants, hydrogen peroxide, free chlorine and monochloramine in groundwater. Results showed that the addition of oxidants did not enhance attenuation of ICMs, since fluence-based decay apparent rate constants were similar (KUV = 3.2 U+00D7 10-3, KUV-Cl2 = 3.6 U+00D7 10-3 and KUV-NH2 = 3.4 U+00D7 10-3 10-3 cm2 mJ-1). This yielded direct photolysis is the main mechanism to attenuate target ICMs.

Drinking water contains organic matter that occasionally needs to be treated to assure its sufficient quality and safety for the consumers. H2O2 and UV advanced oxidation processes (H2O2/UV AOPs) were combined with hydrodynamic cavitation (HC) to assess the effects on the removal of selected organic pollutants. Water samples containing humic acid, methylene blue dye and micropollutants (metaldehyde, diatrizoic acid, iohexol) were treated first by H2O2 (dosages from 1 to 12 mg L-1) and UV (dosages from 300 to 2800 mJ cm-2) AOPs alone and later in combination with HC, generated by nozzles and orifice plates (4, 8, 18 orifices). Using HC, the removal of humic acid was enhanced by 5-15 %, methylene blue by 5-20 % and metaldehyde by approx. 10 %. Under favoring conditions, i.e. high UV absorbance of the matrix (more than 0.050 cm-1 at a wavelength of 254 nm) and a high pollutant to oxidants ratio, HC was found to improve the hydrodynamic conditions in the photolytic reactor, to improve the subjection of the H2O2 to the UV fluence rate distribution and to enhance the removal of the tested organic pollutants, thus showing promising potential of further research in this field.

The aquatic environment becomes increasingly contaminated by anthropogenic pollutants such as pharmaceutical residues. Due to poor biodegradation and continuous discharge of persistent compounds in sewage water samples, pharmaceutical residues might end up in surface waters when not removed. To minimize this pollution, onsite wastewater treatment techniques might complement conventional waste water treatment plants (WWTPs). Advanced oxidation processes are useful techniques, since reactive oxygen species (ROS) are used for the degradation of unwanted medicine residues. In this paper we have studied the advanced oxidation in a controlled laboratory setting using thermal plasma and UV/H2O2 treatment. Five different matrixes, Milli-Q water, tap water, synthetic urine, diluted urine and synthetic sewage water were spiked with 14 pharmaceuticals with a concentration of 5μg/L. All compounds were reduced or completely decomposed by both 150 W thermal plasma and UV/H2O2 treatment. Addnl., also hospital sewage water was tested. First the concentrations of 10 pharmaceutical residues were determined by liquid chromatog. mass spectrometry (LC-MS/MS). The pharmaceutical concentration ranged from 0.08 up to 2400μg/L. With the application of 150 W thermal plasma or UV/H2O2, it was found that overall pharmaceutical degradation in hospital sewage water were nearly equivalent to the results obtained in the synthetic sewage water. However, based on the chem. abatement kinetics it was demonstrated that the degree of degradation decreases with increasing matrix complexity. Since reactive oxygen and nitrogen species (RONS) are continuously produced, thermal plasma treatment has the advantage over UV/H2O2 treatment.

The anthropogenic entry of organic micropollutants into the aquatic environment leads to a potential risk for drinking water resources and the drinking water itself. Therefore, sensitive screening anal. methods are needed to monitor the raw and drinking water quality continuously. Non-target screening anal. has been shown to allow for a more comprehensive investigation of drinking water processes compared to target anal. alone. However, non-target screening is challenging due to the many features that can be detected. Thus, data processing techniques to reduce the high number of features are necessary, and prioritization techniques are important to find the features of interest for identification, as identification of unknown substances is challenging as well. In this study, a drinking water production process, where drinking water is supplied by a water reservoir, was investigated. Since the water reservoir provides surface water, which is anthropogenically influenced by wastewater treatment plant (WWTP) effluents, substances originating from WWTP effluents and reaching the drinking water were investigated, because this indicates that they cannot be removed by the drinking water production process. For this purpose, ultra-performance liquid chromatog. coupled with an ion-mobility high-resolution mass spectrometer (UPLC-IM-HRMS) was used in a combined approach including target, suspect and non-target screening anal. to identify known and unknown substances. Addnl., the role of ion-mobility-derived collision cross sections (CCS) in identification is discussed. To that end, six samples (two WWTP effluent samples, a surface water sample that received the effluents, a raw water sample from a downstream water reservoir, a process sample and the drinking water) were analyzed. Pos. findings for a total of 60 substances in at least one sample were obtained through quant. screening. Sixty-five percent (15 out of 23) of the identified substances in the drinking water sample were pharmaceuticals and transformation products of pharmaceuticals. Using suspect screening, further 33 substances were tentatively identified in one or more samples, where for 19 of these substances, CCS values could be compared with CCS values from the literature, which supported the tentative identification. Eight substances were identified by reference standards In the non-target screening, a total of ten features detected in all six samples were prioritized, whereby metoprolol acid/atenolol acid (a transformation product of the two β-blockers metoprolol and atenolol) and 1,3-benzothiazol-2-sulfonic acid (a transformation product of the vulcanization accelerator 2-mercaptobenzothiazole) were identified with reference standards Overall, this study demonstrates the added value of a comprehensive water monitoring approach based on UPLC-IM-HRMS anal.

Elimination half-life is an important pharmacokinetic parameter that determines exposure duration to approach steady state of drugs and regulates drug administration. The exptl. evaluation of half-life is time-consuming and costly. Thus, it is attractive to build an accurate prediction model for half-life. In this study, several machine learning methods, including gradient boosting machine (GBM), support vector regressions (RBF-SVR and Linear-SVR), local lazy regression (LLR), SA, SR, and GP, were employed to build high-quality prediction models. Two strategies of building consensus models were explored to improve the accuracy of prediction. Moreover, the applicability domains (ADs) of the models were determined by using the distance-based threshold. Among seven individual models, GBM showed the best performance (R2 = 0.820 and RMSE = 0.555 for the test set), and Linear-SVR produced the inferior prediction accuracy (R2 = 0.738 and RMSE = 0.672). The use of distance-based ADs effectively determined the scope of QSAR models. However, the consensus models by combing the individual models could not improve the prediction performance. Some essential descriptors relevant to half-life were identified and analyzed. An accurate prediction model for elimination half-life was built by GBM, which was superior to the reference model (R2 = 0.723 and RMSE = 0.698). Encouraged by the promising results, we expect that the GBM model for elimination half-life would have potential applications for the early pharmacokinetic evaluations, and provide guidance for designing drug candidates with favorable in vivo exposure profile. The morphinan skeleton is an important sub-structure in many medicines such as dextromethorphan, and can be constructed from 1-benzyl-1,2,3,4,5,6,7,8-octahydroisoquinoline (1-benzyl-OHIQ) derivatives 1-Benzyl-3,4,5,6,7,8-hexahydroisoquinolines (1-benzyl-HHIQs), the precursors of 1-benzyl-OHIQs, constitute a type of bulky α, β-unsaturated imines. Until now, the application of imine reductases (IREDs) to α, β-unsaturated imines has only rarely been reported. In this study, through evaluation of 48 IREDs, both enantiomers of 1-(4-methoxybenzyl)-1,2,3,4,5,6,7,8-octahydroisoquinoline (1-(4-methoxybenzyl)-OHIQ) were obtained in high yield and excellent optical purity. Among the enzymes, the most steric hindrance-tolerant IRED from Sandarearacinus amylolyticus (IR40) was able to convert various Ph substituted 1-benzyl-HHIQ to the corresponding 1-benzyl-OHIQ derivatives with excellent enantiomeric excess. These results provide an effective route to synthesize these important compounds via enantioselective reduction of bulky α, β-unsaturated imine precursors, which can be readily prepared from 2-(1-cyclohexenyl)ethylamine and corresponding aryl acetic acids.

In this respect, all of the newly synthesized quinazolinone derivatives I displayed vasorelaxant properties on the isolated thoracic rat aorta. The IC50 of compounds I (R = Ph) (-6.00 ± 0.55), I (R = 4-nitrophenylamino) (-7.31 ± 0.94), I (R = 2,4-dinitrophenylamino) (-7.15 ± 0.81) and I (R = (diphenylmethylidene)aminyl) (-7.77 ± 0.31) was comparable to that seen in the acetylcholine (-7.13 ± 0.14).

The closed basin of the Lower Jordan Valley with the Dead Sea as final sink features high evapotranspiration rates and almost complete reuse of treated wastewater for irrigation farming. This study focuses on the water transfer schemes and the presence, spreading, and potential accumulation of pharmaceutical residues in the local water resources based on findings of a five-year monitoring program. Overall 16 pharmaceuticals and 9 iodinated X-ray contrast media were monitored in groundwater, surface water, and treated wastewater. A total of 95 samples were taken to cover all geog. settings and flow paths from origin (wastewater) to target (groundwater). Nine substances were detected in groundwater, with concentrations ranging between 11 ng/L and 33,000 ng/L. Sometimes, detection rates were higher than in comparable studies: Diatrizoic acid 75%, iopamidol 42%, iopromide 19%, iomeprol 11%, carbamazepine and iohexol 8%, ibuprofen 6%, and fenofibrate and iothalamic acid 3%. Concentrations in groundwater generally increase from north to south depending on the application of treated wastewater for irrigation. Almost all substances occurred most frequently and with highest concentrations in treated wastewater, followed by surface water and groundwater. As exception, diatrizoic acid was found more frequently in groundwater than in treated wastewater, with concentrations being similar. This indicates the persistence of diatrizoic acid with long residence times in local groundwater systems, but may also reflect changing prescription patterns, which would be in accordance with increasing iopamidol findings and surveys at local hospitals. Trend analyses confirm this finding and indicate a high probability of increasing iopamidol concentrations, while other substances did not reveal any trends. However, no proof of evaporative enrichment could be found. The high spatial and temporal variability of the concentrations measured calls for further systematic studies to assess the long-term evolution of organic trace substances in this reuse setting.

Atm. liquefaction technol. has been used widely and is an effective way of biomass component utilization. In this paper, the liquefied products obtained from corn stalk using polyhydric alcs. 1,2-propanediol (PG) mixed diethylene glycol (DEG) through acid catalysis under atm. pressure were characterized by various anal. technologies. The results indicated that 39 kinds of organic compounds were present in bio-oil, among which alcs. were the most, phenols were the second, and their relative contents were 70.7% and 25.6%, resp. There were also some organic acids, ethers, esters, and ketones. More than 80% of these compounds had a carbon number less than 25. Carbon NMR spectra (13C-NMR) showed that different chem. shifts δ (ppm) corresponded to various carbon types. The chem. composition of the residue from liquefaction was complex and contained a certain amount of large mol. substances that were difficult to degrade. It required more severe pyrolysis conditions than those of corn stalk. Results from X-Ray Diffraction (XRD) indicated the destruction of crystalline structure of carbohydrates and the cellulose mols. were cracked, indicating that the cellulose was degraded and the degree of liquefaction was high.
