A three-component coupling of isoquinolines, dimethyl acetylenedicarboxylate (DMAD) and indoles is achieved for the first time to produce dimethyl (E)-2-[1-(1H-3-indolyl)-1,2-dihydro-2-isoquinolinyl]-2-butenedioates in excellent yields and with high selectivity. The reaction proceeds smoothly at room temperature without a catalyst. Quinoline, DMAD and indole also undergo smooth coupling to furnish dimethyl (E)-2-[2-(1H-3-indolyl)-1,2-dihydro-1-quinolinyl]-2-butenedioate under similar conditions. This method is very useful to functionalize both indoles and aza-aromatic compounds in a one-pot operation.

In vitro, compound 26 is able to inhibit exogenous uPA in human chronic wound fluid (IC50 = 0.89 muM). In vivo, in a porcine acute excisional wound model, following topical delivery, compound 26 is able to penetrate into pig wounds and inhibit exogenous uPA activity with no adverse effect on wound healing parameters. On the basis of this profile, compound 26 (UK-371,804) was selected as a candidate for further preclinical evaluation for the treatment of chronic dermal ulcers.

Photocycloaddition and Rearrangement Reactions in a Putative Route to the Skeleton of Plicamine-Type Alkaloids

Two isoquinolones were prepared, to which an allenyl side chain was linked at position C4 via a stereogenic silyloxy-substituted carbon atom. Intramolecular [2+2] photocycloaddition reactions of these substrates proceeded with high diastereoselectivity and delivered the respective cyclobutanes with an exocyclic methylene group (83% and 49% yield). With the 5,6-dioxoloisoquinolone precursor an unprecedented meta-photocycloaddition was observed as a significant side reaction, which occurred at positions C4 and C8a of the isoquinolone skeleton. The cyclobutane products were, after N-alkylation and transformation into the respective cyclobutanones (22-57%), subjected to various rearrangement reactions. In detail, a direct photochemical rearrangement, thermal and photochemical Beckmann rearrangements, and Baeyer-Villiger oxidation reactions were studied. In all cases, products were found, which resulted from cleavage of the amino-substituted cyclobutane bond, but not from the desired cleavage of the alternative alkyl-substituted cyclobutane bond.

Oxidative cyclization of 1-(pyridin-2-yl)guanidine derivatives: A synthesis of [1,2,4]triazolo[1,5-a]pyridin-2-amines and an unexpected synthesis of [1,2,4]triazolo[4,3-a]pyridin-3-amines

Oxidative cyclization of 1-(pyridin-2-yl)guanidine derivatives using N-chlorosuccinimide and aqueous potassium carbonate has been investigated. Chlorination of 1-(5-nitropyridin-2-yl)guanidine by N-chlorosuccinimide in methanol followed by addition of aqueous potassium carbonate gave rise to cyclization and afforded 6-nitro-[1,2,4]triazolo[1,5-a]pyridin-2-amine in one-pot. In the course of studying the scope and limitation of the reaction, it was found that some of the examined 1-(pyridin-2-yl)guanidine derivatives gave not only the desired [1,2,4]triazolo[1,5-a]pyridin-2-amines but also unexpected [1,2,4]triazolo[4,3-a]pyridin-3-amine products. As plausible reaction mechanisms of this oxidative cyclization, diazirine formation and nitrene formation are presented.
