This report describes the discovery of uracil 20, a potent (38 nM in THP1 cells assay) caspase-1 inhibitor for the topical treatment of inflammatory acne. The uracil series was designed according to a published caspase-1 pharmacophore model involving a reactive warhead in P1 for covalent reversible inhibition and an aryl moiety in P4 for selectivity against the apoptotic caspases. Reversibility was assessed in an enzymatic dilution assay or by using different substrate concentrations. In addition to classical structure-activity-relationship exploration, topical administration challenges such as phototoxicity, organic and aqueous solubility, chemical stability in solution, and skin metabolic stability are discussed and successfully resolved.

Efficient saturated red light-emitting polyfluorenes containing iridium complexes in side chains

A series of electrophosphorescent copolymers were synthesized by Suzuki polymerization. A diketone-ended alkyl chain which is grafted in the N-position of the carbazole unit is used as a ligand to form a pendant cyclometalated Ir complex with 1-(9,9-dioctyl-9H-fluoren-2-yl)isoquinoline and 1-(N,N-diphenyl-benzenamine-4-yl)isoquinoline. The electroluminescence from the backbone of the copolymers is completely quenched by tethered iridium complexes even though with the content of iridium complexes being as low as 1 mol%. Saturated red light-emitting diodes were fabricated on the basis of these two series of copolymers. The best device performance with a maximum external quantum efficiency of 7.8% on the basis of copolymer PF-IrNiq1 was achieved, which was among the highest efficiencies of the reported electrophosphorescent red light-emitting polymers comprising iridium ligands.

Synthesis and evaluation of novel chloropyridazine derivatives as potent human rhinovirus (HRV) capsid-binding inhibitors

Human rhinovirus (HRV) is the most important etiologic agent causing common colds. No effective anti-HRV agents are currently available. In this paper we describe the synthesis and the evaluation of novel chloropyridazine derivatives (compounds 5a-g) as potent human rhinovirus (HRV) capsid-binding inhibitors. Results showed that compound 5e and 5f exhibited effective anti-HRV activity against HRV-2 and HRV-14. In addition, compound 5e and 5f showed lower cytotoxicity than Pirodavir.

A complex phenomenon was observed in the process of preparing hydroxyisoquinoline through copper-catalyzed hydrolysis of bromoisoquinoline. The copper (II) complexes of hydroxyisoquinoline (L2Cu.5H2O) were characterized by high resolution mass spectra, thermogravimetric analysis, IR, 1H nuclear magnetic resonance (NMR), and 2D-NMR. The Cu (II) complexes were mononuclear and coordinated with oxygen and nitrogen atom of two hydroxyisoquinoline and five water molecules in which a strong hydrogen bond was present. Two optimized methods had been studied to prevent the formation of copper (II) complexes. The isoquinoline with 4, 5, 6, 7, and 8 hydroxyl substitutions were successfully prepared by copper-catalyzed hydrolysis of corresponding bromoisoquinoline and then workup by sodium sulfide or adjusted pH by dry ice or carbon dioxide gas.
