Some of these product structures are found in various naturally occurring and biologically active heterocyclic compounds. The reaction involves an unusual multiple construction of new C – C, C – N, and C – H bonds and the cleavage of a C – H bond in one pot. It is likely that the three-component coupling proceeds through the nucleophilic addition of quinoline to benzyne, which generates a zwitterionic species. The latter then attracts a proton from terminal alkyne (or ketone) to generate an N-arylated quinolinium cation and an acetylide anion. Further reaction of these two ions provides the final substituted 1,2-dihydroquinolines. In the reaction, the terminal alkyne acts first as a proton donor and then as a nucleophile. The application of a three-component coupling reaction product, 1,2-dihydro-2-pyridinyl alkyne in a stereospecific [4+2] Diels-Alder cycloaddition reaction with N-phenyl maleimide to give an isoquinuclidine derivative, an important core present in various natural products, is demonstrated.

A series of alpha,beta-unsaturated hydroxamic acid derivatives as novel HDAC inhibitors (HDACi) with structural modifications of the connecting unit and the CAP group was synthesized. The in vitro evaluation against the human cancer cell lines A2780 and Cal27 identified 6e and 7j as the most potent compounds regarding HDAC inhibitory activity and inhibition of proliferation. Isoform profiling against HDAC2, 4, 6 and 8 revealed a preference for HDAC2 and 6 for both compounds in contrast to the pan HDACi panobinostat. 6e and 7j enhanced significantly cisplatin-induced cytotoxicity in a combination treatment mediated by increased apoptosis induction and caspase-3/7 activation. The interaction between 6e or 7j and cisplatin was highly synergistic and more pronounced for the cisplatin resistant subline Cal27CisR. IC50 values of cisplatin were even lower in Cal27CisR pretreated with 6e or 7j than for the parental cell line Cal27. Based on our findings, the novel dual class I/HDAC6 inhibitors could serve as an option to overcome cisplatin resistance with fewer side effects in comparison to panobinostat.

Analogues of the potent alpha2-adrenoceptor antagonist (8aR,12aS,13aS)-5,8,8a,9,10,11,12,12a,13,13a-decahydro-3-methoxy-12- (methylsulfonyl)-6H-isoquino[2,1-g][1,6]naphthyridine (1b) were prepared and evaluated for alpha1- and alpha2-adrenoceptor affinity. Affinity for [3H]yohimbine-labeled alpha2-adrenoceptors was found to be highly dependent on the stereochemistry of the tetracyclic system. The 8abeta,12aalpha,13aalpha diastereomer of 1 (56) had moderate affinity for alpha2-adrenoceptors while the 8abeta,12abeta,13aalpha diastereomer (55) had very low affinity. The affinity and selectivity of these agents for alpha2-adrenoceptors was found to correspond to that observed for several isomeric yohimbine analogues which have similar relative and absolute stereochemistries. Deviation from the structure of 1 by opening the B ring, changing the position of the sulfonamide nitrogen, or changing the attachment of the D ring led to a dramatic decrease in alpha2-adrenoceptor affinity. High binding affinity was found to correlate with functional antagonism in the guinea pig ileum. This multistage process involves double allylmetallation of the heterocyclic ring followed by cyclization through intramolecular carbometallation of the C=C bond. The structures of three key derivatives of 1,4-ethano-2,3-dihydroisoquinoline were established by X-ray diffraction analysis. The reactions of quinoxaline with allyl-and methallylzinc bromide proceeded stereospecifically to form trans-2,3-diallylated 1,2,3,4-tetrahydroquinoxalines. Heating of quinoline with methallylzinc bromide in THF afforded 4-methallylquinoline in nearly quantitative yield. The initially formed 1,4-metallation product underwent aromatization through elimination of HZnBr.

Effect of ethyl groups at two different positions on the electronic, optical and charge transport properties of 5,5-difluoro-1,37,9-tetramethyloctahydro-1H,5H-514-dipyrrolo[1,2-c:2?,1?-f] [1?3] diazaborinine (Comp_1) as parent molecule has been studied at molecular level. The introduction of ethyl at two positions in Comp_1 led to a red shifted absorption spectra in comparison with parent molecule in the violet region. Various properties of interest such as highest occupied molecular orbitals (HOMO), lowest unoccupied molecular orbitals (LUMO), vertical electron affinity (EAv), adiabatic electron affinity (EAa), vertical ionization potential (IPv), adiabatic ionization potential (IPa), electron reorganization energies (lambdae) and hole reorganization energies (lambdah) were explored.
