OsVI(N)(L)2Cl (L = 2-quinaldinate) (2a) reacts also with [PPN](N3) to give an osmium(III) dichloro complex, trans-[PPN][OsIII(L)2Cl2] (6). Reaction of OsVI(N)(L)2Cl (L = 2-quinaldinate) (2a) with lithium sulfide produces an osmium(II) thionitrosyl complex OsII(NS)(L)2Cl (7). We report a direct C-H to C-C bond functionalization of electron-deficient heteroaromatics enabled by mild C-C bond cleavage of cyclopropanols as a new route to beta-aryl carbonyl-containing products. Additionally, as an alternative to using a “catalyst” that requires an excess amount of a sacrificial oxidant for regeneration and/or oxidative aromatization, this paper features manganese(IV) oxide as an inexpensive “dual role” reagent – effecting both C-C bond cleavage and ultimate rearomatization. Under the specified conditions, a variety of heterocycles proved competent for regioselective C-C bond formation, alongside a diverse array of cyclopropanols with broad functional-group tolerance. We highlight applications to complex-molecule synthesis and direct derivatization of biologically active alkaloids. Furthermore, kinetic isotope effect (KIE) experiments, radical scavengers, and some insight into the application of tri- and tetravalent manganese species are invoked to shape an initial mechanistic hypothesis.

On the basis of the spectroscopic and photochemical evidence, N – O bond scission is the dominant primary photochemical process involving the CT state, the subsequent radical coupling resulting in efficient aromatic hydroxylation. The outcome of both quenching and sensitization experiments confirms this assertion. The results strongly suggest that the ensuing photohydroxylation reaction is not a concerted process, but rather a two-step N – O bond scission followed by C – O bond formation, which is regioselectively guided by the electronic distribution of the resulting donor cation-radical.

The synthesis of a new family of octahedral Ir(iii) complexes with dual cyclometalating phosphine chelates, namely: 1-(diphenylphosphino)naphthalene (dpnaH) and isoquinoline (dppiH), is reported. The inhibition of CYP17 to block androgen biosynthesis is a well validated strategy for the treatment of prostate cancer. Herein we reported the design, synthesis and structure?activity relationship (SAR) study for a series of novel 1,2,3,4- tetrahydrobenzo[4,5]thieno[2,3-c]pyridine derivatives. Some analogs demonstrated a potent inhibition to both rat and human CYP17 protein and reduced testosterone production in human H295R cell line. Some analogs also showed high selectivity against other CYP enzymes such as 3A4, 1A2, 2C9, 2C19 and 2D6, which may limit side effects due to drug-drug interactions. Among these analogs, the most potent compound 9c showed 1.5 fold more potent against rat and human CYP17 protein than that of abiraterone (IC50 = 16 nM and 20 nM vs. 25 nM and 36 nM respectively). In NCI-H295R cells, the inhibitory effect of compound 9c on testosterone production (52± 2%) was also more potent than that of abiraterone (74± 15%) at the concentration of 1 muM. Further, it was shown that 9c reduced plasma testosterone level in a dose-dependent manner in Sprague-Dawley rats. Thus, analog 9c maybe a potential agent used for the treatment of prostate cancer.
