Sensing smell is a highly complex biol. process, and characterizing and mimicking the interaction between the olfactory receptor (OR) protein and its ligands is extremely challenging. Herein, we report a highly sensitive and selective human nose-like nanobioelectronic nose (nbe-nose), which responds to gaseous odorants sensitively and selectively, has a signal specificity pattern similar to that in the cellular signal transduction pathway, and maintains an antagonistic behavior similar to the human nose. The human olfaction mechanism was mimicked by using carboxylated polypyrrole nanotubes (CPNTs) functionalized with human OR protein. The nbe-nose was able to detect gaseous odorants at a concentration as low as 0.02 parts-per-trillion (ppt), which was comparable to a highly trained, human expert’s nose. The nbe-nose can be used scientifically for smell mechanism studies. It can be also applied to various fields that rely on smell monitoring for industrial and public purposes.

An interaction of odorants with olfactory receptors is thought to be the initial step in odorant detection. However, ligands have been reported for only 6 out of 380 human olfactory receptors, with their structural determinants of odorant recognition just beginning to emerge. Guided by the notion that amino acid positions that interact with specific odorants would be conserved in orthologs, but variable in paralogs, and based on the prediction of a set of 22 of such amino acid positions, we have combined site-directed mutagenesis, rhodopsin-based homol. modeling, and functional expression in HeLa/Olf cells of receptors OR1A1 and OR1A2. We found that (i) their odorant profiles are centered around citronellic terpenoid structures, (ii) two evolutionary conserved amino acid residues in transmembrane domain 3 are necessary for the responsiveness of OR1A1 and the mouse ortholog Olfr43 to (S)-(-)-citronellol, (iii) changes at these two positions are sufficient to account for the differential (S)-(-)-citronellol responsiveness of the paralogs OR1A1 and OR1A2, and (iv) the interaction sites for (S)-(-)-citronellal and (S)-(-)-citronellol differ in both human receptors. Our results show that the orientation of odorants within a homol. modeling-derived binding pocket of olfactory receptor orthologs is defined by evolutionary conserved amino acid positions.

Recent studies have shown that sperm chemotaxis critically involves the human olfactory receptor OR1D2, which is activated by the aromatic aldehyde bourgeonal. Given that both natural and sexual selection may act upon the expression of receptors, we hypothesized that human males are more sensitive than human females for bourgeonal. Using a 3-alternative forced-choice test procedure, olfactory detection thresholds were determined for a total of 500 subjects, 250 males, and 250 females between 18 and 40 years of age. We found that male subjects detected bourgeonal at significantly lower concentrations (mean value: 13 ppb) compared with female subjects (mean value: 26 ppb), whereas no such gender difference in olfactory sensitivity was found with helional, a structural analog of bourgeonal, and with n-pentyl acetate, an aliphatic ester, which were tested in parallel. Males and females did not differ in their frequency of specific anosmia for any of the 3 odorants. The frequency distributions of olfactory detection thresholds were monomodal with all 3 odorants in both genders. Olfactory detection thresholds did not differ significantly between pre- and postovulatory females with any of the 3 odorants. To the best of our knowledge, this is the first study ever to find a human male superiority in olfactory sensitivity. Single nucleotide polymorphisms and/or copy number variations in genes coding for olfactory receptors may be the proximate cause for our finding, whereas a gender difference in the behavioral relevance of bourgeonal may be the ultimate cause.

We have measured concentration detection (i.e., psychometric) functions to determine the odor detectability of homologous aliphatic aldehydes (propanal, butanal, hexanal, octanal, and nonanal) and helional. Subjects (16 �n �18) used a 3-alternative forced-choice procedure against carbon-filtered air (blanks), under an ascending concentration approach. Generation, delivery, and control of each vapor were achieved via an 8-station vapor delivery device. Gas chromatog. served to quantify the concentrations presented. Group and individual functions were modeled by a sigmoid (logistic) equation. Odor detection thresholds (ODTs) were defined as the concentration producing a detectability (P) halfway (P = 0.5) between chance (P = 0.0) and perfect detection (P = 1.0). ODTs decreased with carbon chain length: 2.0, 0.46, 0.33, and 0.17 ppb, resp., from propanal to octanal, but the threshold increased for nonanal (0.53 ppb), revealing maximum sensitivity for the 8-carbon member. The strong olfactory receptor (OR) ligands octanal and helional (0.14 ppb) showed the lowest thresholds. ODTs fell at the lower end of previously reported values. Interindividual variability (ODT ratios) amounted to a factor ranging from 10 to 50, lower than typically reported, and was highest for octanal and hexanal. The behavioral dose-response functions emerge at concentrations 2-5 orders of magnitude lower than those required for functions tracing the activation of specific human ORs by the same aldehydes in cell/mol. studies, after all functions were expressed as vapor concentrations

Humans are able to detect and discriminate myriads of odorants using only several hundred olfactory receptors (ORs) classified in two major phylogenetic classes representing ORs from aquatic (class I) and terrestrial animals (class II). Olfactory perception results in a combinatorial code, in which one OR recognizes multiple odorants and different odorants are recognized by different combinations of ORs. Moreover, recent data suggest that odorants could also behave as antagonists for other ORs, thus making the combinatorial coding more complex. Here we describe the odorant repertoires of two human ORs belonging to class I and class II, resp. For this purpose, we set up an assay based on calcium imaging in which 100 odorants were screened using air-phase odorant stimulation at physiol. doses. We showed that the human class I OR52D1 is functional, exhibiting a narrow repertoire related to that of its orthologous murine OR, demonstrating than this human class I OR is not an evolutionary relic. The class II OR1G1 was revealed to be broadly tuned towards odorants of 9-10 carbon chain length, with diverse functional groups. The existence of antagonist odorants for the class II OR was also demonstrated. They are structurally related to the agonists, with shorter carbon chain length.

The detection of chem. distinct odorants starts with the binding of an odorant ligand to a specific receptor protein on the olfactory neuron cell surface. To address the problem of olfactory perception at a mol. level, we have expressed and characterized different olfactory receptors with several expression systems. Here we provide the first documentation of functional expression of odorant receptors using the Semliki Forest virus system. The human odorant OR 17-40 receptor and the rat 17 receptor were functionally expressed in vertebrate kidney cells (HEK293) using recombinant Semliki Forest viruses. Receptors were expressed as a fusion protein with the N-terminal membrane import sequence of the guinea pig serotonin receptor. Experiments employing the Ca2+-sensitive dye fura-2 revealed a fast, transient increase in the [Ca2+]i after application of the specific agonists helional and octanal to HEK293 cells infected with viruses containing RNA for the human odorant OR 17-40 receptor and the rat 17 receptor, resp.

Regarding the chemotaxis of sperms, new insights have been gained during the last 20 years. Olfactory receptors are expressed on the flagellar midpiece of human spermatogenic cells. One of them, OR1D2, is also expressed in the olfactory epithelium. This receptor has been suggested to play a role in sperm chemotaxis and thus in fertility. As OR1D2 is activated by bourgeonal, the aim of the study was to investigate whether patients with idiopathic infertility would exhibit a decreased olfactory sensitivity toward bourgeonal. Participants were 14 patients with idiopathic infertility and 23 controls (all young fathers). After having ascertained normosmia, odor thresholds and intensity ratings for the pleasant and flowery odors of bourgeonal, helional, and phenylethylalc. were obtained. As a result, patients had specifically decreased intensity ratings for bourgeonal. It suggests that men with unexplained infertility tend to be less suprathreshold sensitive toward the odor of bourgeonal but not to that of other floral odors. It may be speculated that the decreased olfactory sensitivity relates to a decreased functionality of OR1D2, which in turn may be linked to idiopathic infertility.

The authors describe how mammalian olfactory receptors (ORs) could be used as sensing elements of highly specific and sensitive bioelectronic noses. An OR and an appropriate Gα protein were co-expressed in Saccharomyces cerevisiae cells from which membrane nanosomes were prepared, and immobilized on a sensor chip. By Surface Plasmon Resonance, the authors were able to quant. evaluate OR stimulation by an odorant, and G protein activation. The authors demonstrate that ORs in nanosomes discriminate between odorant ligands and unrelated odorants, as in whole cells. This assay also provides the possibility for quant. assessment of the coupling efficiency of the OR with different Gα subunits, without the interference of the cellular transduction pathway. The authors’ findings will be useful to develop a new generation of electronic noses for detection and discrimination of volatile compounds, particularly amenable to micro- and nano-sensor formats.
