A wide range of aromatic aldehydes can smoothly undergo the three-component cyclization reaction under the optimized conditions to give the corresponding products in moderate to good yields.

In this study 14 novel hydrazide-hydrazones of 5-bromo-2-iodobenzoic acid (3-16) were synthesized on the basis of condensation reaction. The chem. structure of obtained derivatives was established on the basis of spectral data (1H NMR and 13C NMR) and the lipophilicity of synthesized mols. was determined with the use of RP-HPTLC chromatog. Synthesized hydrazide-hydrazones (3-16) were subjected to in vitro cytotoxicity assay and antimicrobial activity anal. against a panel of bacteria and fungi. Among newly synthesized derivatives (3-16), compound 5 was characterized by high, selective and the most diverse cytotoxicity to the cancer cell lines. Mols. 7 and 9 which were substituted with a nitro group in the Ph ring also exhibited very significant inhibitory effect in the tumor cells and they were very selective. Similarly, compound 13 showed high antiproliferative activity against both cancer cell lines (769-P, HepG2) with satisfactory selectivity. In turn, mol. 8 was characterized by lower inhibitory effect in tumor cells but high selectivity. Derivative 16 proved to be toxic mainly to 769-P cells plausibly by the inhibition of COX-2 mediated signalling pathway. In summary, the introduction of chloro substituent or nitro group to the mol. proved to be most advantageous, providing high cytotoxicity and selectivity to tumor cells. However, the presence of indole scaffold appeared to be responsible for COX-2 inhibitory effect. Some of synthesized hydrazide-hydrazones possessed also moderate antimicrobial activity against a panel of microorganisms.

A series of 4-(quinoline-4-amino) benzoylhydrazide derivatives I (R = Ph, 3-bromo-4-methoxyphenyl, 3-bromothiophene-2-yl, 2-chloropyridin-3-yl, etc.) was synthesized and evaluated for their anti-HCC activity and binding affinity to Nur77 in vitro. Compound I (R = pyridin-4-yl) emerged as the best Nur77 binder (KD = 1.17μM) and has potentially selective cytotoxicity to HCC cells. Mechanistically, I (R = pyridin-4-yl) extensively induced caspase-independent cytoplasmic vacuolization and paraptosis through Nur77-mediated ER stress and autophagy. Moreover, I (R = pyridin-4-yl) exhibited an effective xenograft tumor inhibition by modulating Nur77-dependent cytoplasmic vacuolation and paraptosis. This paper is the first to disclose that chemotherapeutic agents targeting Nur77-mediated cytoplasmic vacuolization and paraptosis may provide a promising strategy to combat HCC that frequently evade the apoptosis program.

In a continuing effort to develop new CDK9 inhibitors endowed with good anticancer activity, a series of new benzene carbohydrazide derivatives I (R = pyridin-2-yl, pyridin-3-yl, pyridin-4-yl; R 1 = n-Pr, 4-methylphenyl, thiophen-2-yl, etc.) bearing a (pyridyl pyrimidin-2-yl)amino moiety in position-4 of the Ph ring was designed and synthesized. This work reports the preparation of benzene carbohydrazide derivatives I, their inhibition effect on HIV-1 transcription, and the preliminary structure-activity relationships. Compound I (R = pyridin-2-yl, R 1 = 4-methoxyphenyl (II)) was found to be the most potent CDK9 inhibitor and exhibited excellent anti-proliferative activities against cancer cells (A375, A549, HepG2, and MCF-7) but was less toxic to normal cells (MCF-10A and HaCaT). Further bioassays indicated that compound II could induce the apoptosis of cancer cells, which contributes to its antitumor effects. Finally, the mol. docking studies are performed to predict the binding mode of 9h at the ATP binding site of CDK9 and identify the essential amino acids responsible for the interactions.

A series of (alkyl/aryl)kojylmethane (IKM) derivatives I [R = 4-HOC6H4, 2-pyrrolyl, 3-indolyl, etc.; R1 = n-Bu, Ph, 2-naphthyl, etc.] was synthesized using a multicomponent one-pot reaction under solvent-free condition using a heterogeneous catalyst. The synthesized compounds were screened against breast cancer cell lines MDA-MB-231, MCF7, and T47D. The structure-activity relationship revealed that IKM synthesized from aliphatic aldehydes were active against all three cell lines and showed IC50 value of 0.15μM-3.93μM. IKM synthesized from aromatic aldehydes having electron-donating groups specifically inhibited the proliferation of the MDA-MB-231 cell line. The effect of various substituted indoles was also studied and observed that compound I [R = 5-CN-indol-3-yl, R1 = Ph] synthesized from 5-cyanoindole, specifically inhibited T47D cell line proliferation. Compound I [R = 3-indolyl, R1 = n-heptyl] synthesized from kojic acid, indole and octanal was found to be most potent with IC50 values of 0.21, 0.15, and 3.45μM against MDA-MB-231, MCF7, and T47D, resp.
