N-Myristoyltransferase (NMT) represents a promising drug target for human African trypanosomiasis (HAT), which is caused by the parasitic protozoa Trypanosoma brucei. We report the optimization of a high throughput screening hit (1) to give a lead mol. DDD85646 (63), which has potent activity against the enzyme (IC50 = 2 nM) and T. brucei (EC50 = 2 nM) in culture. The compound has good oral pharmacokinetics and cures rodent models of peripheral HAT infection. This compound provides an excellent tool for validation of T. brucei NMT as a drug target for HAT as well as a valuable lead for further optimization.

Moreover, it’s suggested that low water solubility of chems. might induce false neg. outcomes. Thus, in this study, we tested relatively low water soluble 37 chems. with log Kow values above and below 3.5 in the h-CLAT. The small-scale assessment resulted in nine false neg. outcomes for chems. with log Kow values greater than 3.5. We then created a dataset of 143 chems. by combining the existing dataset of 106 chems. and examined the predictive performance of the h-CLAT for chems. with a log Kow of less than 3.5; a total of 112 chems. from the 143 chems. in the dataset. The sensitivity and overall accuracy for the 143 chems. were 83% and 80%, resp. In contrast, sensitivity and overall accuracy for the 112 chems. with log Kow values below 3.5 improved to 94% and 88%, resp. These data suggested that the h-CLAT could successfully detect sensitizers with log Kow values up to 3.5. When chems. with log Kow values greater than 3.5 that were deemed pos. by h-CLAT were included with the 112 chems., the sensitivity and accuracy in terms of the resulting applicable 128 chems. out of the 143 chems. became 95% and 88%, resp. The use of log Kow values gave the h-CLAT a higher predictive performance. The aim of the study is efficacy and potential benefits of cyclodextrins in delivering fragrances with and without CDs were compared using headspace/solid-phase microextraction gas chromatog. All measurements and assessments were carried out on the volar forearms(s) of one Caucasian subject in a controlled environment: 25 ± 1°C and 30% ± 5% relative humidity. Imines or Schiff bases (SB) are formed by the condensation of an aldehyde or a ketone with a primary amine, with the removal of a water mol. Schiff bases are central mols. in several biol. processes for their ability to form and cleave by small variation of the medium. We report here the controlled hydrolysis of four SBs that may be applied in the fragrance industry, as they are profragrances all containing odorant mols.: Me anthranilate as primary amine, and four aldehydes (cyclamal, helional, hydroxycitronellal and triplal) that are very volatile odorants. The SB stability was assessed over time by HPLC-MS in neutral or acidic conditions, both in solution and when trapped in low mol. weight gels. Our results demonstrate that it is possible to control the hydrolysis of the Schiff bases in the gel environment, thus tuning the quantity of aldehyde released and the persistency of the fragrance.

Single atom catalysts (SACs) possess ultimate atom utilization efficiency and outstanding catalytic performance. Herein, we synthesize Zn SACs (Zn-TpPa) anchored on a COF (TpPa-1) using a facile solution method. Catalyzed by Zn-TpPa, CO2 and N-methylamine are transformed into N-methylformanilide under mild reaction conditions with a TOF of 17,155 h-1, which is the highest among all reported recyclable Zn-based catalysts. Zn-TpPa can also catalyze N-formylation of many other amines with excellent yields. The higher reactivity was attributable to the well-dispersed Zn active sites on COF and outstanding adsorption of CO2 owing to high surface area of COF. Our research provides a facile method for constructing SACs as well as an effective pathway for CO2 transformation and environmental protection.

Meta-anal. of In the United States, cancer is the second leading cause of mortality, and millions more battle cancer worldwide. As such, primary prevention of cancer is a major interest globally. Aspirin has been studied as a primary prevention method for multiple diseases, mainly cardiovascular disease and various forms of cancer. The role of aspirin as a primary prevention of cancer is still controversial and may be more beneficial in certain cancers over others. With rapidly surfacing large randomized controlled trials (RCTs) studying this subject, we aimed to evaluate the efficacy and safety of aspirin as a primary prophylaxis for cancer. A comprehensive electronic database search was conducted for all RCTs that compared aspirin vs. placebo for the prevention of any type of disease, and where cancer incidence or mortality was reported. The primary outcome was cancer related mortality. Secondary outcomes were cancer incidence, all-cause mortality, major bleeding, any bleeding and gastrointestinal (GI) bleeding. We calculated risk ratios (RRs) and 95% confidence intervals (CIs) using a random-effects model at the longest follow-up period. We included 16 RCTs with 104,018 total patients, mean age of 60.51 years, mean follow-up of 5.48 years, and a male percentage of 38.72%. We found that aspirin was not associated with a significant reduction of cancer-related mortality compared with placebo (RR 0.99; 95% CI: 0.87-1.12; P = 0.85: I2 = 41%). Compared with placebo, aspirin was not associated with significant reduction of all-cause mortality (RR 0.97; 95% CI: 0.92-1.02; P = 0.19; I2 = 13%) or cancer incidence (RR: 0.98; 95% CI: 0.92-1.04; P = 0.43; I2 = 16%). However, aspirin treatment was associated with significantly increased risks of any bleeding (RR 1.63; 95% CI: 1.31-2.03; P < 0.01), major bleeding (RR 1.41; 95% CI: 1.26-1.57; P < 0.01), and GI bleeding (RR 1.85; 95% CI: 1.38-2.48; P < 0.01) compared with placebo. Our study did not find any significant reductions in cancer-related mortality or cancer incidence when compared aspirin use with placebo or no aspirin. Our study also highlights that the use of aspirin for primary prevention of cancer was found to cause higher rates of bleeding (any bleeding, major bleeding, and GI bleeding) compared to placebo or no aspirin at the longest follow-up period with no significant benefit in cancer primary prevention.
