The action of ClO· is more prominent than HO· and Cl· in the advanced oxidation degradation of some pollutants. However, studies on the pollutant degradation mechanism and kinetics by ClO· are limited. In this study, 43 different kind of aromatic compounds which are important anthropogenic and natural water pollutants were selected as models to investigate their ClO· oxidation mechanism and kinetics computationally. The results showed that radical adduct formation (RAF) rather than single electron transfer (SET) reaction was prominent in ClO·-initiated reactions of aromatic compounds In subsequent reactions of the ClO-adduct, the Cl end of the -OCl moiety shifted to the benzene ring, which was the key to hydroxylation and chlorination of aromatic compounds by ClO·. The calculated ClO· initiated reaction rate constants (k·ClO) of aromatic compounds were 102-1010 M-1 s-1. ClO· was highly reactive to phenolates, anilines and alkoxy/hydroxyl aromatic compounds Upon the deprotonation of phenol to phenolate, the k·ClO value increased by 4 orders of magnitude. The k·ClO values of alkoxybenzenes were higher for compounds with shorter alkyl side chains and more alkoxy substituents. The k·ClO increased for anilines with longer alkyl side chains. The k·ClO quant. structure-activity relationships (QSARs) models were developed to predict the reactivity of ClO· to complex aromatic compounds in aquatic systems.

Background/Aims: Notably, activating transcriptional factor 2 (ATF2), a histone-modification gene, is involved in oxidative stress-induced apoptosis. The aim of this study was to clarify the role of ATF2 in contrast media-induced nephropathy. Methods: Human embryonic kidney 293T cells were treated with four different contrast media:ionic high-osmolar diatrizoate; ionic low-osmolar iothalamate; non-ionic low-osmolar iohexol, and non-ionic iso-osmolar iodixanol. The mRNA expression of ATF2 was determined by real-time PCR. Short interfering RNA was used to knock down ATF2 mRNA expression. Phosphorylation of ATF2 was measured by Western blotting. Wistar rats were administered either diatrizoate or a normal saline injection. Apoptosis in kidney tubular cells was determined by the presence of pos. TUNEL stain. Results: Diatrizoate, iodixanol and iothalamate, but not iohexol, induced the expression of ATF2 mRNA and phosphorylation of ATF2 in 293T cells in a time-dependent manner. More apoptotic cells were in diatrizoate-treated kidney cells than in the saline injection group (p < 0.00001). Cell death was significantly increased by knockdown ATF2 expression in the presence of diatrizoate, indicating a protective role of ATF2 in contrast media-induced apoptosis. Conclusions: Differential activation of ATF2 by different contrast media provides a new insight into the mechanism and prevention of contrast-induced nephropathy. Insect olfactory receptors are heteromeric ligand-gated ion channels composed of � common subunit (Orco) and � subunit that confers odorant specificity. Little is known about how individual subunits contribute to the structure and function of the olfactory receptor complex. We expressed insect olfactory receptors in Xenopus oocytes to investigate 2 functional features, ion channel block and odorant recognition. The sensitivity of Drosophila olfactory receptors to inhibition by ruthenium red, a cation channel blocker, varied widely when different specificity subunits were present, suggesting that the specificity subunits contribute to the structure of the ion pore. Olfactory receptors formed by DmelOr35a and Orco subunits from several different species displayed highly similar odorant response profiles, suggesting that the Orco subunit does not contribute to the structure of the odorant-binding site. We further explored odorant recognition by conducting a detailed examination of the odorant specificity DmelOr67a + DmelOrco, a receptor that responds to aromatic structures. This screen identified agonists, partial agonists, and an antagonist of DmelOr67a + DmelOrco. Our findings favor specific subunit arrangements within the olfactory receptor complex and provide a preliminary odorophore for an olfactory receptor, offering a useful foundation for future exploration of insect olfactory receptor structure.

Biol. treatment processes have the potential to remove organic micropollutants (OMPs) duringwater treatment. The OMP removal capacity of conventional drinking water treatment processes such asrapid sand filters (RSFs), however, has not been studied in detail. We investigated OMP removal and transformation product (TP) formation in seven full-scale RSFs all treating surface water, using highresoln. mass spectrometry based quant. suspect and non-target screening (NTS). Addnl.,we studied the microbial communities with 16S rRNA gene amplicon sequencing (NGS) in both influent and effluent waters as well as the filter medium, and integrated these data to comprehensively assess the processes that affect OMP removal. In the RSF influent, 9 to 30 of the 127 target OMPs were detected. Theremoval efficiencies ranged from 0 to 93%. A data-driven workflow was established to monitor TPs, basedon the combination of NTS feature intensity profiles between influent and effluent samples and the prediction of biotic TPs. The workflow identified 10 TPs, including mol. structure. Microbial community composition anal. showed similar community composition in the influent and effluent of most RSFs, but different from the filter medium, implying that specific microorganisms proliferate in the RSFs. Some of these are able to perform typical processes in water treatment such as nitrification and iron oxidation

Fibromyalgia (FM) is characterized by chronic widespread pain. Here, we probed the metabolomics profiles of serum and urine in a mouse model of FM induced by intermittent cold stress (ICS). We focused on the role of acid-sensing ion channel 3 (ASIC3) in the metabolomics profiling because ICS treatment induced chronic widespread muscle pain lasting for 1 mo in wild-type (Asic3+/+) but not Asic3-knockout (Asic3-/-) mice. Serum and urine samples were collected from both genotypes at different ICS stages, including before ICS (basal level) and post-ICS at days 10 (middle phase, P10) and 40 (recovery phase, P40). Finally, we developed an algorithm for the impactful metabolites in the FM model including cis-aconitate, kynurenate, taurine, pyroglutamic acid, pyrrolidonecarboxylic acid, and 4-methoxyphenylacetic acid in urine as well as carnitine, deoxycholic acid, lysoPC(16:0), lysoPC(20:3), oleoyl-L-carnitine, and trimethylamine N-oxide in serum. Asic3-/- mice were impaired in only muscle allodynia development but not other pain symptoms in the ICS model, so the ASIC3-dependent metabolomics changes could be useful for developing diagnostic biomarkers specific to chronic widespread muscle pain, the core symptom of FM. Further pharmacol. validations are needed to validate these metabolomics changes as potential biomarkers for FM diagnosis and/or treatment responses.

The field of heterogeneous photoredox catalysis has grown substantially and impacted organic synthesis because of the affordability and reusability of catalysts. This study reports radical trifluoromethylation with Cd-chalcogenide semiconductors. Cd semiconductors, particularly CdSe, are readily available, com., visible-light-responsive, heterogeneous photocatalysts. The potential of readily available Cd semiconductors, particularly CdSe, is confirmed by their increased photocatalytic activity toward trifluoromethylation with various substrates, such as (hetero)arenes and vinylic amides/acids, via addition, cyclization, and decarboxylation under visible light. The economic significance of this strategy is also highlighted through the scalable synthesis of biol. active mols. followed by catalyst reuse. Moreover, these catalysts are relatively inexpensive compared with transition metal-based homogeneous photocatalysts, presently used in organic synthesis.

Stereoselective synthesis of trisubstituted alkenes is a long-standing challenge in organic chem., due to the small energy differences between E and Z isomers of trisubstituted alkenes (compared with 1,2-disubstituted alkenes). Transition metal-catalyzed isomerization of 1,1-disubstituted alkenes can serve as an alternative approach to trisubstituted alkenes, but it remains underdeveloped owing to issues relating to reaction efficiency and stereoselectivity. Here we show that a novel cobalt catalyst can overcome these challenges to provide an efficient and stereoselective access to a broad range of trisubstituted alkenes. This protocol is compatible with both mono- and dienes and exhibits a good functional group tolerance and scalability. Moreover, it has proven to be a useful tool to construct organic luminophores and a deuterated trisubstituted alkene. A preliminary study of the mechanism suggests that a cobalt-hydride pathway is involved in the reaction. The high stereoselectivity of the reaction is attributed to both a π-π stacking effect and the steric hindrance between substrate and catalyst.

Although hybrid bidentate ligands are known to yield highly enantioselective products in asym. hydrogenation (AH), synthesis of these ligands is an arduous process. Herein, a one pot, atom-economic synthesis of a hybrid phosphine-phosphite (L1) is reported. After understanding the reactivity difference between an O-nucleophile vs. C-nucleophile, one pot synthesis of Senphos (L1) was achieved (72%). When L1 was treated with [Rh], 31P NMR revealed bidentate coordination to Rh. Senphos, in the presence of rhodium, catalyzes the AH of Methyl-2-acetamido-3-phenylacrylate and discloses an unprecedented turn over frequency of 2289, along with excellent enantio-selectivity (92%). The generality is demonstrated by hydrogenating an array of alkenes. The AH operates under mild conditions of 1-2 bar H2 pressure, at room temperature The practical relevance of L1 is demonstrated by scaling-up the reaction to 1 g and by synthesizing DOPA, a drug widely employed for the treatment of Parkinson’s disease. Computational insights indicate that the R isomer is preferred by 3.8 kcal/mol over the S isomer.
