The structure, electronic properties, vibrational fundamental modes, thermodynamic properties and chemical shifts of the compounds are investigated. The vibrational fundamental modes are assigned and analysed by utilising FT?IR and FT?Raman spectrum of the compounds recorded in the range of 4000?400 and 4000?100 cm?1, respectively. The LUMO?HOMO energy difference is determined to explain the charge transfer within the molecule. The molecular electrostatic potential and total electron density of the molecules are determined to explain the size, shape and the reactive sites (polar) of the molecules. The 1H and 13C NMR chemical shifts of the compounds are determined by Gauge Invariant Atomic Orbital (GIAO) method using DMSO?d6 solvent by B3LYP/cc?pVTZ method. The local and global chemical reactivity properties of the compounds have been investigated with the help of global and local reactivity descriptors.

By linking the iron(iii)-binding siderophore azotochelin to an iridium-containing imine-reduction catalyst that produces racemic product in the absence of the protein CeuE, but a reproducible enantiomeric excess if protein bound, the assembly and reductively triggered disassembly of the ATHase was achieved. The crystal structure of the ATHase identified the residues involved in high-affinity binding and enantioselectivity. While in the presence of iron(iii), the azotochelin-based anchor binds CeuE with high affinity, and the reduction of the coordinated iron(iii) to iron(ii) triggers its dissociation from the protein. Thus, the assembly of the artificial enzyme can be controlled via the iron oxidation state.

Metal-Binding Pharmacophore Library Yields the Discovery of a Glyoxalase 1 Inhibitor

Anxiety and depression are common, highly comorbid psychiatric diseases that account for a large proportion of worldwide medical disability. Glyoxalase 1 (GLO1) has been identified as a possible target for the treatment of anxiety and depression. GLO1 is a Zn2+-dependent enzyme that isomerizes a hemithioacetal, formed from glutathione and methylglyoxal, to a lactic acid thioester. To develop active inhibitors of GLO1, fragment-based drug discovery was used to identify fragments that could serve as core scaffolds for lead development. After screening a focused library of metal-binding pharmacophores, 8-(methylsulfonylamino)quinoline (8-MSQ) was identified as a hit. Through computational modeling and synthetic elaboration, a potent GLO1 inhibitor was developed with a novel sulfonamide core pharmacophore. A lead compound was demonstrated to penetrate the blood-brain barrier, elevate levels of methylglyoxal in the brain, and reduce depression-like behavior in mice. These findings provide the basis for GLO1 inhibitors to treat depression and related psychiatric illnesses.

Three analogue series were designed and synthesized to improve the bioavailability profile of UCPH-102, but none displayed substantially improved properties in this respect. In vitro profiling of UCPH-102 (10 mum) at 51 central nervous system targets in radioligand binding assays strongly suggests that the compound is completely selective for EAAT1. Finally, in a rodent locomotor model, p.o. administration of UCPH-102 (20 mg kg-1) did not induce acute effects or any visible changes in behavior. EAAT1 inhibition beyond the BBB: In the present study, oral administration (40 mg kg-1) of the selective excitatory amino acid transporter subtype 1 (EAAT1) inhibitor UCPH-102 in rats yielded respective plasma and brain concentrations of 10.5 and 6.67 mum after 1 h. Methyl 2-furoate: An alternative reagent to furan for palladium-catalysed direct arylation

The palladium-catalysed direct arylation of methyl 2-furoate with aryl bromides was studied. The use of KOAc as the base, dimethylacetamide (DMAc) as the solvent and Pd(OAc)2 as the catalyst was found to give 5-arylfurans regioselectively and without decarboxylation. These methyl 5-aryl-2-furoates gave 2,5-diarylfurans by decarboxylative coupling by using Pd(OAc)2 as the catalyst. Methyl 2-furoate thus represents a convenient alternative substrate to furan for the synthesis of mono- or poly-arylated furans.
