Synthetic investigations of (1,3?)-bistetrahydroisoquinolines are reported as the key intermediates for the synthesis of ecteinascidin and phthalascidin pentacyclic structure analogues through successive Pictet-Spengler cyclization and intramolecular peptide coupling. The direct Pictet-Spengler reaction between a derivative of l-DOPA and N-protected-alpha-aminoaldehyde was first extended to the synthesis of cis-(1,3?)- bistetrahydroisoquinoline. After introduction of the required amino acid moiety, an efficient six-membered ring intramolecular peptide coupling gave rise to piperazine derivative structures. Complete structural assignments were corroborated by NMR and X-ray spectroscopic methods. Nevertheless, the optical integrity of the N-protected-alpha-aminoaldehyde seems to be sensitive to the reaction conditions. Pentacylic structures, having an anti C3-C11 backbone stereochemistry, were obtained from cyclization para- and ortho- to the 3-OH group of the l-DOPA derivative.

CONSTRUCTION OF A DNA-PROBE TO DETECT ISOQUINOLINE-DEGRADING BACTERIA

To facilitate the cloning of DNA encoding isoquinoline degradation an assay was developed that allowed the rapid visual scoring of the isoquinoline degradation phenotype of single colonies. Transposon mutagenesis of one of the isolates, Comamonas acidovorans IQ3, was performed using Tn5, and nine Isq(-) mutants deficient in the ability to utilise isoquinoline as the sole nitrogen source were isolated. These mutants were also incapable of utilising the first metabolite of the isoquinoline degradation pathway, 1-hydroxyisoquinoline, as the sole carbon source. For each Isq(-) mutant, the EcoRI fragment containing the Tn5 insertion was cloned into pBR322. Restriction and Southern analyses of the cloned DNA revealed that of the nine Isq(-) mutants, six contained Tn5 insertions in a common 8.9-kb EcoRI fragment derived from the wild type, C. acidovorans IQ3. The cloned DNA thought to be involved in the degradation of isoquinoline proved to be specific when used as a probe in colony hybridization to some bacteria possessing the ability to degrade isoquinoline.

Decomposition of isoquinoline and quinoline by supercritical water

The ability of supercritical water (SCW) to decompose heterocyclic compounds (quinoline and isoquinoline) has been explored in this study. The results obtained suggest that water acts as a chemical reagent above its critical point (374 degrees C and 22.1 MPa). Significant proportions of isoquinoline and quinoline were removed during the reaction with SCW. The response of these compounds to pyrolysis was also compared with their reaction with SCW. Both compounds were relatively more reactive in the presence of SCW than during pyrolysis. Because of the different positions of N atom in the two compounds, they reacted with SCW differently.
