Three novel chromophores B-D have been synthesized based on thiophene derivatived donors, including 5-(methyl(phenyl)amino)thiophene 5-(bis(4-methoxyphenyl)amino)thiophene 5-((4-methoxyphenyl)(methyl)amino)thiophene groups resp., with the same isophorone-derived bridge and tricyanovinyldihydrofuran (TCF) acceptors. A new strategy was proposed to improve the electron donating ability of the chromophore, thienyl groups were explored as a replacement for the Ph unit in the donor part of the chromophore. D. functional theory calculations suggested that the first hyperpolarizability of (arylamino) thiophene donor-containing chromophore B-D is about 20% higher than that of 4-(dialkylamino) Ph counterpart chromophore A. The syntheses of the benzo-bridged dioxa- and diazatriangulenium derivatives (BDOTA+ and BDATA+, resp.) required two different synthetic approaches, which reflect the structural and physiochem. impact on the reactivity of [5]-helicenium precursors. Spectroscopic investigations show that introduction of the benzo-bridge into the triangulenium chromophore significantly red shifts the absorption and emission while maintaining fluorescence lifetimes above 10 ns. The combination of high quantum yield, long fluorescence lifetime, and emission above 600 nm is possible only if the structural aspects of the triangulenium framework are perfectly harmonized to secure a low rate of non-radiative deactivation. The new benzo-bridge may be a general motif to obtain red shifted derivatives of other dye classes.

Sesquiterpenoids were found in high concentrations in the oils of E. patrisii and M. splendens, which were rich in E-caryophyllene (32.0% and 45.8%); E. stipitata and M. sylvatica, which displayed germacrene D (11.8%) and germacrene B (24.5%); and P. guajava that showed epi-β-bisabolol (16.1%) as the main compound However, P. guineense samples (Pgui-1 and Pgui-2) were rich in monoterpenoids such as limonene (Pgui-1: 30.2%; Pgui-2 30.4%) and α-pinene (Pgui-1: 22.5%; Pgui-2: 17.7%). The samples showed a weak and moderate antioxidant activities in the DPPH assay, displaying inhibition rates from 11.5% to 38.6% (at 10 mg/mL). All samples were cytotoxic against human cancer cells by the MTT method. Epat oil showed higher activity against melanoma (SKMEL-19, IC505.8μg/mL), gastric (AGP01, IC503.2μg/mL), and colon (HCT116, IC506.7μg/mL). In recent years, the work of various research groups has allowed the substitution of the hazardous solvents most widely used in solid-phase peptide synthesis, namely DMF, NMP, DCM, DEE, among others, by several much less hazardous solvents. The higher b.p. of these solvents compared with the DCM allows the preparation of protected peptides with less risk of premature removal of the most labile protecting groups, such as the Trt of His. Our findings once again evidence the value/versatility of green solvents in strict chem. terms.

We report the on-demand generation of hexane-soluble (2-ethylhexyl)sodium (1) from 3-(chloromethyl)heptane (2) using a sodium-packed-bed reactor under continuous flow conditions. Thus, the resulting solution of 1 is free of elemental sodium and therefore suited for a range of synthetic applications. This new procedure avoids the storage of an alkylsodium and limits the handling of metallic sodium to a min. (2-Ethylhexyl)sodium (1) proved to be a very useful reagent and undergoes in-line Br/Na-exchanges as well as directed sodiations. The resulting arylsodium intermediates are subsequently trapped in batch with various electrophiles such as ketones, aldehydes, Weinreb-amides, imines, allyl bromides, disulfides and alkyl iodides. A reaction scale-up of the Br/Na-exchange using an in-line electrophile quench was also reported.

A turn-on two-photon fluorescent probe HCA-Green (I) for hypochlorous acid (HOCl) was synthesized using 4-methylamino-1,8-naphthalimide (MNA) as a two-photon fluorophore and p-hydroxyaniline as a leaving-recognition domain. Both the probe and the fluorophore were studied under one- and two-photon excitation modes. The fluorescence intensity of the probe was enhanced by �29-fold and �93-fold under one-photon and two-photon excitation, resp., after reacting with HOCl. A maximal two-photon action cross-section of 50 GM was obtained under excitation at 810 nm. The probe exhibited high sensitivity with a detection limit of 42.3 nM, as well as high selectivity, low cytotoxicity, and good photostability. Two-photon microscopy (TPM) was conducted to visualize HOCl levels in living cells and tissues. The production of endogenous HOCl induced by lipopolysaccharide-mediated inflammation was successfully monitored with this probe.
