Mirabile, Salvatore published the artcileEvaluation of 4-(4-Fluorobenzyl)piperazin-1-yl]-Based Compounds as Competitive Tyrosinase Inhibitors Endowed with Antimelanogenic Effects, Formula: C9H10O3, the main research area is fluorobenzylpiperazinyl derivative tyrosinase inhibitor antimelanogenic; Docking studies; Homology model; Kinetic mechanism; Synthesis; Tyrosinase inhibitors.

There is a considerable attention for the development of inhibitors of tyrosinase (TYR) as therapeutic strategy for the treatment of hyperpigmentation disorders in humans. Continuing in our efforts to identify TYR inhibitors, we describe the design, synthesis and pharmacophore exploration of new small mols. structurally characterized by the presence of the 4-fluorobenzylpiperazine moiety as key pharmacophoric feature for the inhibition of TYR from Agaricus bisporus (AbTYR). Our investigations resulted in the discovery of the competitive inhibitor [4-(4-fluorobenzyl)piperazin-1-yl]-(3-chloro-2-nitro-phenyl)methanone 26 (I, IC50=0.18μM) that proved to be ∼100-fold more active than reference compound kojic acid (IC50=17.76μM). Notably, compound 26 exerted antimelanogenic effect on B16F10 cells in absence of cytotoxicity. Docking anal. suggested its binding mode into AbTYR and into modelled human TYR.

Mast cells (MCs) play key roles in IgE-mediated immunoresponses, including in the protection against parasitic infections and the onset and/or symptoms of allergic diseases. IgE-mediated activation induces MCs to release mediators, including histamine and leukotriene, as an early response, and to produce cytokines as a late phase response. Attempts have been made to identify novel antiallergic compounds from natural materials such as Chinese medicines and food ingredients. We herein screened approx. 60 compounds and identified salicylaldehyde, an aromatic aldehyde isolated from plant essential oils, as an inhibitor of the IgE-mediated activation of MCs. A degranulation assay, flow cytometric analyses, and enzyme-linked immunosorbent assays revealed that salicylaldehyde inhibited the IgE-mediated degranulation and cytokine expression of bone-marrow-derived MCs (BMMCs). The salicylaldehyde treatment reduced the surface expression level of FcεRI, the high affinity receptor for IgE, on BMMCs, and suppressed the IgE-induced phosphorylation of tyrosine residues in intercellular proteins, possibly Lyn, Syk, and Fyn, in BMMCs. We also examined the effects of salicylaldehyde in vivo using passive anaphylaxis mouse models and found that salicylaldehyde administration significantly enhanced the recovery of a reduced body temperature due to systemic anaphylaxis and markedly suppressed ear swelling, footpad swelling, and vascular permeability in cutaneous anaphylaxis.

Cai, Jing published the artcileCorrelation Analysis of Microbiota and Volatile Flavor Compounds of Caishiji Soybean Paste, HPLC of Formula: 21834-92-4, the main research area is volatile flavor compound microbiota soybean paste.

Microbial diversity plays a crucial part in the fermentation of Caishiji soybean paste (CSP). In the current study, the microbiota and volatile flavor compounds (VFCs) in CSP were identified through Illumina MiSeq sequencing and headspace gas chromatog.-mass spectrometry. Five bacterial (Bacillus, Tetragenococcus, Salinivibrio, Halomonas, and Staphylococcus) and four fungal genera (Aspergillus, Debaryomyces, Nigrospora, and Curvularia) were revealed as dominant among the entire microbiome of CSP. More than 70 VFCs, including 8 acids, 15 esters, 8 alcs., 14 aldehydes, 4 ketones, 5 phenols, and 20 miscellaneous VFCs were detected during the fermentation process. A total of 12 kinds of VFCs were identified in the odor activity value (OAV) anal. The results of the correlation anal. between microbiota and VFCs indicated that Bacillus, Tetragenococcus, Staphylococcus, and Aspergillus were the main microbiota affecting the flavor of CSP. These results may serve as a reference for enhancing the quality of CSP.

Owusu, Margaret’s team published research in Journal of Food Science and Technology (New Delhi, India) in 2013 | CAS: 21834-92-4

Journal of Food Science and Technology (New Delhi, India) published new progress about Astringency. 21834-92-4 belongs to class isoquinoline, name is 5-Methyl-2-phenylhex-2-enal, and the molecular formula is C13H16O, Application In Synthesis of 21834-92-4.

Owusu, Margaret published the artcileRelationship of sensory and instrumental aroma measurements of dark chocolate as influenced by fermentation method, roasting and conching conditions, Application In Synthesis of 21834-92-4, the main research area is aroma flavor fermentation roasting conching dark chocolate; Chocolate; Conching; Flavor attributes; Heap-fermented; Roasting; Tray-fermented.

The investigation was undertaken to establish a relationship between key odorants and perceived flavor attributes of dark chocolate as influenced by cocoa fermentation method, roasting and conching conditions, using multivariate data anal. in an attempt to use one variable to predict the other. Eight of the sixteen flavor attributes used by a trained sensory panel to describe and quantify the intensity of attributes in the samples were significantly different (p<0.05). Roasting significantly reduced astringency in heap-fermented samples but the reduction in tray-fermented samples was not significant. Unconched samples were rated higher in banana attribute than conched samples. Multivariate data anal. tools, Principal Component Anal. (PCA) and Partial Least Squares (PLS) were used to investigate quant. descriptive anal. and GC-O data and also to relate the two sets of data. PLS 1 models based on single sensory attributes gave better models than PLS2 models based on all sixteen sensory attributes. Ethyl-3-methylbutanoate (fruity, flowery); 2,5-dimethylpyrazine (popcorn); dihydro-2(3H)-furanone, (sweet); linalool oxide (sweet, flowery); benzaldehyde (earthy, nutty) and 2/3-methylbutanal (cocoa, roasted) modeled fruit attribute. It was also possible to model the attribute astringent from the aroma compounds 5-methyl-2-phenyl-2-hexenal (sweet, roasted cocoa), ethyl-3-methylbutanoate and pentyl acetate (green, cucumber). Since fruit attribute was higher in unconched samples and astringent higher in unroasted samples, it may be possible to use the levels of these important aroma compounds as indicators of the sensory attributes fruit and astringent. Lee, Sang Hun published the artcileMimicking the human smell sensing mechanism with an artificial nose platform, Synthetic Route of 1205-17-0, the main research area is odor sensor olfactory sensor olfaction carboxylated polypyrrole nanotube electrode.

Sensing smell is a highly complex biol. process, and characterizing and mimicking the interaction between the olfactory receptor (OR) protein and its ligands is extremely challenging. Herein, we report a highly sensitive and selective human nose-like nanobioelectronic nose (nbe-nose), which responds to gaseous odorants sensitively and selectively, has a signal specificity pattern similar to that in the cellular signal transduction pathway, and maintains an antagonistic behavior similar to the human nose. The human olfaction mechanism was mimicked by using carboxylated polypyrrole nanotubes (CPNTs) functionalized with human OR protein. The nbe-nose was able to detect gaseous odorants at a concentration as low as 0.02 parts-per-trillion (ppt), which was comparable to a highly trained, human expert’s nose. The nbe-nose can be used scientifically for smell mechanism studies. It can be also applied to various fields that rely on smell monitoring for industrial and public purposes.

Musa, Musiliyu A. published the artcileIdentification of 7,8-dihydroxy-3-phenylcoumarin as a reversible monoamine oxidase enzyme inhibitor, Application of 4-Methoxyphenylacetic acid, the main research area is dihydroxy phenylcoumarin monoamine oxidase enzyme inhibitor neuroprotection; 3-arylcoumarin; molecular modeling; monoamine oxidase inhibitors; neuroprotection; structure-activity relationship.

We herein report the biol. evaluation of 3-arylcoumarin derivatives (3a-l) as potential human monoamine oxidase-A and -B (hMAO-A and hMAO-B) inhibitors. The result indicated that 7,8-dihydroxy-3-(4-nitrophenyl)coumarin (3j) was most effective against MAO-A (inhibition concentration [IC50] = 6.46 ± 0.02μM) and MAO-B (IC50 = 3.8 ± 0.3μM) enzymes than other synthesized compounds and reference compounds (pargyline and moclobemide). Furthermore, compound (3j) showed (a) nonselectivity against hMAO enzymes, (b) reversible hMAO enzymes inhibition, and (c) neuroprotection against H2O2-treated human neuroblastoma (N2a) cells. Finally, a mol. modeling study revealed that the hMAO enzymes inhibitory activity of the compound (3j) may be due to the orientation where the nitro (NO2) group lies deep into the receptor and the Ph ring directed toward flavin adenosine dinucleotide via hydrogen bond interaction, and possible π-π interaction with various important residues. Thus, the results of the present study demonstrate that compound (3j) can be considered as a promising scaffold for the development of hMAO-A and hMAO-B inhibitors.

Schmiedeberg, Kristin published the artcileStructural determinants of odorant recognition by the human olfactory receptors OR1A1 and OR1A2, COA of Formula: C11H12O3, the main research area is olfactory receptor OR1A1 A2 gene odorant.

An interaction of odorants with olfactory receptors is thought to be the initial step in odorant detection. However, ligands have been reported for only 6 out of 380 human olfactory receptors, with their structural determinants of odorant recognition just beginning to emerge. Guided by the notion that amino acid positions that interact with specific odorants would be conserved in orthologs, but variable in paralogs, and based on the prediction of a set of 22 of such amino acid positions, we have combined site-directed mutagenesis, rhodopsin-based homol. modeling, and functional expression in HeLa/Olf cells of receptors OR1A1 and OR1A2. We found that (i) their odorant profiles are centered around citronellic terpenoid structures, (ii) two evolutionary conserved amino acid residues in transmembrane domain 3 are necessary for the responsiveness of OR1A1 and the mouse ortholog Olfr43 to (S)-(-)-citronellol, (iii) changes at these two positions are sufficient to account for the differential (S)-(-)-citronellol responsiveness of the paralogs OR1A1 and OR1A2, and (iv) the interaction sites for (S)-(-)-citronellal and (S)-(-)-citronellol differ in both human receptors. Our results show that the orientation of odorants within a homol. modeling-derived binding pocket of olfactory receptor orthologs is defined by evolutionary conserved amino acid positions.

Olsson, Peter published the artcileHuman Male Superiority in Olfactory Sensitivity to the Sperm Attractant Odorant Bourgeonal, HPLC of Formula: 1205-17-0, the main research area is sperm attractant odorant bourgeonal olfactory system gender olfaction.

Recent studies have shown that sperm chemotaxis critically involves the human olfactory receptor OR1D2, which is activated by the aromatic aldehyde bourgeonal. Given that both natural and sexual selection may act upon the expression of receptors, we hypothesized that human males are more sensitive than human females for bourgeonal. Using a 3-alternative forced-choice test procedure, olfactory detection thresholds were determined for a total of 500 subjects, 250 males, and 250 females between 18 and 40 years of age. We found that male subjects detected bourgeonal at significantly lower concentrations (mean value: 13 ppb) compared with female subjects (mean value: 26 ppb), whereas no such gender difference in olfactory sensitivity was found with helional, a structural analog of bourgeonal, and with n-pentyl acetate, an aliphatic ester, which were tested in parallel. Males and females did not differ in their frequency of specific anosmia for any of the 3 odorants. The frequency distributions of olfactory detection thresholds were monomodal with all 3 odorants in both genders. Olfactory detection thresholds did not differ significantly between pre- and postovulatory females with any of the 3 odorants. To the best of our knowledge, this is the first study ever to find a human male superiority in olfactory sensitivity. Single nucleotide polymorphisms and/or copy number variations in genes coding for olfactory receptors may be the proximate cause for our finding, whereas a gender difference in the behavioral relevance of bourgeonal may be the ultimate cause.
