Critical to the success of this process are not only the use of the copper catalyst as a dual photo- and cross-coupling catalyst but also tuning of the NHP-type esters to inhibit the facile homodimerization of the alkyl radical and terminal alkyne, resp. Owing to the use of stable and easily available NHP-type esters, the reaction features a broader substrate scope compared with reactions using the alkyl halide counterparts, covering (hetero)benzyl-, allyl-, and aminocarbonyl-substituted carboxylic acid derivatives, and (hetero)aryl and alkyl as well as silyl alkynes, thus providing a vital complementary approach to the previously reported method.

Intriguingly, we found that the dehydroxymethylation process can be significantly promoted by the cerium catalyst, and the stabilization effect of the fragmented radicals also plays a significant role. This operationally simple protocol has enabled the direct utilization of primary alcs. as unconventional alkyl nucleophiles for radical-mediated 1,4-conjugate additions with Michael acceptors. A broad range of alcs., from simple ethanol to complex nucleosides and steroids, have been successfully applied to this fragment coupling transformation. Furthermore, the modularity of this catalytic system has been demonstrated in diversified radical-mediated transformations including hydrogenation, amination, alkenylation, and oxidation

The human gut microbiome is crucial in modulating host health mostly through bacterial metabolites. Chem. exposure is typical external stress which alters its composition and functionality. To date, very few studies have investigated the effect of feeding state on chem.-induced gut microbial metabolic dysregulations. Here, we set up an in vitro human gut microbiome and incorporated a metabolomics approach to investigate the effect of tetracycline (TET) at multiple doses (i.e., 10, 1, and 0.01 mg/L) on gut microbiome under the fed and fasted states. Overall, the metabolome was highly responsive at the fed state with 62 metabolites dysregulated while only 14 were altered at the fasted state under 10 mg/L (clin. TET dose). As expected, nutrient consumption was significantly inhibited under clin. TET dose at the fed state accumulating nutrients such as glutamate and leucine. Interestingly, at the fed state, TET could increase the synthesis of indole and Ph derivatives including indole-3-aldehyde and hydrocinnamate, while inhibiting indoxyl, tryptamine, and vitamin B production, all of which have host health implications. Furthermore, metabolites like indoxyl and xanthurenic acid were still responsive at 0.01 mg/L (dietary TET dose). Collectively, results demonstrated that the feeding state greatly modulates the chem.-induced gut microbial metabolic alterations.

In view of numerous pharmaceutical applications of biscoumarins, the construction of appropriate one-pot synthetic methods for their preparation has been a subject of great attention in recent years. In continuation of ongoing study for the construction of promising approaches for the preparation of biscoumarins, herein we report a zeolite-mediated methodol., in which zeolites, for example, NaY, HY, HZSM-5 and H-beta were employed as catalysts to investigate their catalytic potencies in the synthesis of biscoumarins. In this approach, aromatic aldehydes were treated with 4-hydroxycoumarin in the presence of zeolite catalyst in refluxing ethanol for 2-3 h to furnish biscoumarins in 80-96% yield. The yield of biscoumarins increased in the following order NaY < HZSM-5 < H-beta < HY.Aldehydes both with electron-donating and electron-withdrawing units underwent smooth conversion to their resp. biscoumarins without the development of any side products. The cost-effective, eco-friendly and ready available zeolite catalyst could be recovered and reused several times. This methodol. offers operational simplicity and avoids the use of toxic solvents and corrosive reagents. The present approach offers remarkable advantages such as high catalytic activity, straight-forward procedure, simple workup and low catalyst loading. Those 3-arylcoumarin derivatives were screened for antioxidant, α-glucosidase inhibitory and advanced glycation end-products (AGEs) formation inhibitory. Most compounds exhibited significant antioxidant and AGEs formation inhibitory activities. Anti-diabetic activity studies showed that compounds and were equipotent to the standard drug glibenclamide in vivo. According to the exptl. results, the target compound can be used as a lead compound for the development of new anti-diabetic drugs. The whole experiment showed that anti-diabetic activity is prevalent in 3-arylcoumarins, which added a new natural skeleton to the development of anti-diabetic active drugs.
