Achieving NIR Emission for Donor-Acceptor Type Platinum(II) Complexes by Adjusting Coordination Position with Isomeric Ligands

Four platinum(II) complexes Pt-1, Pt-2, Pt-3, and Pt-4 with the isomeric donor-acceptor (D-A) conjugated ligand framework are designed and prepared, and their thermal, photophysical, and electrochemical characteristics investigated. Crystal structures for Pt-1 and Pt-4 are determined with single-crystal X-ray diffraction analysis, showing distorted and nonplanar geometry. Complex Pt-4 exhibits much greater distortion, attributed to the steric interactions between benzothiadiazole and naphthalene. Remarkably different photophysical, electrochemical, and electroluminescent properties are found for these platinum(II) complexes. Photoluminescence wavelengths of these complexes range from 590 to 800 nm with bandgaps of 1.7-2.0 eV. Coordination with [1,2,5]thiadiazolo[3,4-c]pyridine and triphenylamine can enhance D-A interactions, reducing the bandgap and producing near-infrared emission for Pt-3. Organic light-emitting devices (OLEDs) display electroluminescence with emission peaks at 626, 645, 826, and 571 nm, with maximum external quantum efficiencies of 0.13%, 0.04%, 0.49%, and 0.22% for Pt-1, Pt-2, Pt-3, and Pt-4 doped OLEDs, respectively. Thus, adjusting the coordination position with the isomeric conjugation framework ligand is an appropriate strategy to tune the light-emitting properties of platinum complexes in OLEDs.

Herein, we describe the X-ray structure-based design and optimization of biaryl mannoside FimH inhibitors. Diverse modifications to the biaryl ring to improve druglike physical and pharmacokinetic properties of mannosides were assessed for FimH binding affinity based on their effects on hemagglutination and biofilm formation along with direct FimH binding assays. Substitution on the mannoside phenyl ring ortho to the glycosidic bond results in large potency enhancements several-fold higher than those of corresponding unsubstituted matched pairs and can be rationalized from increased hydrophobic interactions with the FimH hydrophobic ridge (Ile13) or “tyrosine gate” (Tyr137 and Tyr48) also lined by Ile52. The lead mannosides have increased metabolic stability and oral bioavailability as determined from in vitro PAMPA predictive model of cellular permeability and in vivo pharmacokinetic studies in mice, thereby representing advanced preclinical candidates with promising potential as novel therapeutics for the clinical treatment and prevention of recurring urinary tract infections.

A three-component coupling of isoquinolines, dimethyl acetylenedicarboxylate (DMAD) and indoles is achieved for the first time to produce dimethyl (E)-2-[1-(1H-3-indolyl)-1,2-dihydro-2-isoquinolinyl]-2-butenedioates in excellent yields and with high selectivity. The reaction proceeds smoothly at room temperature without a catalyst. Quinoline, DMAD and indole also undergo smooth coupling to furnish dimethyl (E)-2-[2-(1H-3-indolyl)-1,2-dihydro-1-quinolinyl]-2-butenedioate under similar conditions. This method is very useful to functionalize both indoles and aza-aromatic compounds in a one-pot operation.

Palladium(II)-Catalyzed One-Pot Syntheses of 9-(Pyridin-2-yl)-9H-carbazoles through a Tandem C-H Activation/C-X (X=C or N) Formation Process

A new one-pot synthesis of 9-(pyridin-2-yl)-9H-carbazoles through the simultaneous C-H activation and palladium(II)-catalyzed cross-coupling of N-phenylpyridin-2-amines with potassium aryltrifluoroborates is presented. Silver acetate and 1,4-dioxane proved to be the best oxidant and solvent, respectively. The product yields fluctuated from modest to excellent and the reaction showed sufficient functional group tolerance. p-Benzoquinone served as an important ligand for the transmetalation and reductive elimination steps in the catalytic process. The kinetic isotope effects (kH/kD) for the first and second C-H activation/C-C or C-N formation steps were measured as 2.14 and 1.18, respectively. Finally, a rational catalytic mechanism is presented based on all experimental evidence. Thrown together to perform: A variety of N-pyridylcarbazoles were synthesized through concurrent C-H activation/coupling of N-phenylpyridin-2-amines with potassium aryltrifluoroborates (see scheme; BQ=p-benzoquinone). The catalytic mechanism was elucidated by the examination of X-ray crystal structures of PdII intermediates and key products, as well as control experiments and kinetic isotope effect studies. With a dual organocatalytic system involving a chiral phosphoric acid and a dicyanopyrazine-derived chromophore (DPZ) photosensitizer and under the irradiation with visible light, an enantioselective Minisci-type addition of alpha-amino acid-derived redox-active esters (RAEs) to isoquinolines has been developed. A variety of prochiral alpha-aminoalkyl radicals generated from RAEs were successfully introduced on isoquinolines, providing a range of valuable alpha-isoquinoline-substituted chiral secondary amines in high yields with good to excellent enantioselectivities.
