Pharmaceuticals find their way to the aquatic environment via wastewater treatment plants (WWTPs). Biotransformation plays an important role in mitigating environmental risks; however, a mechanistic understanding of involved processes is limited. The aim of this study was to evaluate potential relationships between first-order biotransformation rate constants (kb) of nine pharmaceuticals and initial concentration of the selected compounds, and sampling season of the used activated sludge inocula. Four-day bottle experiments were performed with activated sludge from WWTP Groesbeek (The Netherlands) of two different seasons, summer and winter, spiked with two environmentally relevant concentrations (3 and 30 nM) of pharmaceuticals. Concentrations of the compounds were measured by LC-MS/MS, microbial community composition was assessed by 16S rRNA gene amplicon sequencing, and kb values were calculated The biodegradable pharmaceuticals were acetaminophen, metformin, metoprolol, terbutaline, and phenazone (ranked from high to low biotransformation rates). Carbamazepine, diatrizoic acid, diclofenac, and fluoxetine were not converted. Summer and winter inocula did not show significant differences in microbial community composition, but resulted in a slightly different kb for some pharmaceuticals. Likely microbial activity was responsible instead of community composition In the same inoculum, different kb values were measured, depending on initial concentration In general, biodegradable compounds had a higher kb when the initial concentration was higher. This demonstrates that Michealis-Menten kinetic theory has shortcomings for some pharmaceuticals at low, environmentally relevant concentrations and that the pharmaceutical concentration should be taken into account when measuring the kb in order to reliably predict the fate of pharmaceuticals in the WWTP. Reject water contains complex components of organic compounds, which have significant influences on the nitrogen removal performance when treated using biol. autotrophic nitrogen removal technol. In this study, a two-stage partial nitritation (PN)-anammox (floc-granule) system was established to treat reject water (COD/NH+4-N = 0.97 ± 0.15), and the evolution of organic compounds along PN and annamox bioreactors was investigated using gas chromatog.-mass spectrometry and excitation-emission matrix. Also, the variation of PN and anammox sludge properties relating to COD reduction was examined The PN-anammox system removed approx. 80% of total inorganic nitrogen and COD with hydraulic reaction time of 16 h. The influent organics (330-600 mg COD/L) in reject water were primarily composed of volatile, protein-like and humic acid-like organic compounds PN process contributed 53 ± 18% of the overall COD removal, primarily including oxygen-containing organics (e.g. phenol), proteins and humic acids. Anammox process contributed 22 ± 15% of the overall COD removal, but large mol. acids (e.g. lactic acid) and small mol. alcs. (e.g. glycerol) were reoccurred, contributing to the effluent COD with recalcitrant hydrocarbons (e.g. n-Octadecane). Reject water increased the extracellular proteins/polysaccharides ratio of PN and anammox sludge, promoting the adsorption and degradation of organic compounds High-throughput sequencing results showed that denitrifying bacteria of Ottowia increased from 0.03% to 14.4% in PN reactor, and of Denitratisoma increased from 9.6% to 15.4% in anammox reactor. The occurrence of these denitrifiers might mitigate the neg. impact of organics to functional organisms. This study highlights the organics fate during PN-anammox treatment system, which is important to maintain the robust nitrogen removal when treating organics-containing and high ammonium concentration wastewater.

Abstract: Aberrant activation of ERK signaling pathway usually leads to oncogenesis, and small mol. agents targeting this pathway are impeded by the emergence of drug resistance due to reactivation of ERK signaling. Compound DEL-22379 has been reported to inhibit ERK dimerization which was unaffected by drug-resistant mechanism reactivating the ERK signaling. Here, we discussed a structure-activity relationship study of DEL-22379. Forty-seven analogs were designed and synthesized. Each synthesized compound was biol. evaluated for their inhibitory rates on several tumor cell lines and compounds with high inhibitory rates were further evaluated for IC50 values. The structure-activity relationship of idolin-2-one scaffold and the impact of Z/E configuration on potency were discussed. Potential safety of two synthesized analogs was investigated and in silico docking study of five compounds was performed to understand the structural basis of ERK dimerization inhibition. A total of 81 volatile compounds including 15 hydrocarbons, 10 alcs., 7 acids, 12 aldehydes, 4 esters, 19 S-containing and N-containing compounds, and 14 others were isolated and identified in 1 h-marinated and cooked duck meats. A total of 101 kinds of volatile flavor compounds including 13 hydrocarbons, 14 alc., 7 acids, 8 aldehydes, 12 esters, 23 S-containing and N-containing compounds, and 24 others were detected in 3 h-marinated duck meats of the three body parts. It was proved in this study that under the same conditions, the volatile compounds in duck legs are more than those in duck breasts and wings, and the types of volatile flavor substances increase significantly in duck meats after cooked. Practical applications Prepared and cooked duck meat, especially wings and legs are popular food in China. The results suggest that Pickling makes duck meat more flavory while proper pickling time is less than 3 h. Duck wings are better for marinating and cooking compared with duck breasts and legs. The acceptance study of duck meat from different body parts and the anal. of volatile flavor compounds are beneficial for optimizing utilization of whole parts of duck meat.
