In the year 2010, effluents from 90 European wastewater treatment plants (WWTPs) were analyzed for 156 polar organic chem. contaminants. The analyses were complemented by effect-based monitoring approaches aiming at estrogenicity and dioxin-like toxicity analyzed by in vitro reporter gene bioassays, and yeast and diatom culture acute toxicity optical bioassays. Analyses of organic substances were performed by solid-phase extraction (SPE) or liquid-liquid extraction (LLE) followed by liquid chromatog. tandem mass spectrometry (LC-MS-MS) or gas chromatog. high-resolution mass spectrometry (GC-HRMS). Target microcontaminants were pharmaceuticals and personal care products (PPCPs), veterinary (antibiotic) drugs, perfluoroalkyl substances (PFASs), organophosphate ester flame retardants, pesticides (and some metabolites), industrial chems. such as benzotriazoles (corrosion inhibitors), iodinated x-ray contrast agents, and gadolinium magnetic resonance imaging agents; in addition biol. endpoints were measured. The obtained results show the presence of 125 substances (80% of the target compounds) in European wastewater effluents, in concentrations ranging from low nanograms to milligrams per L. These results allow for an estimation to be made of a European median level for the chems. investigated in WWTP effluents. The most relevant compounds in the effluent waters with the highest median concentration levels were the artificial sweeteners acesulfame and sucralose, benzotriazoles (corrosion inhibitors), several organophosphate ester flame retardants and plasticizers (e.g. tris(2-chloroisopropyl)phosphate; TCPP), pharmaceutical compounds such as carbamazepine, tramadol, telmisartan, venlafaxine, irbesartan, fluconazole, oxazepam, fexofenadine, diclofenac, citalopram, codeine, bisoprolol, eprosartan, the antibiotics trimethoprim, ciprofloxacine, sulfamethoxazole, and clindamycine, the insect repellent N,N’-diethyltoluamide (DEET), the pesticides MCPA and mecoprop, perfluoroalkyl substances (such as PFOS and PFOA), caffeine, and gadolinium.

This study aimed at evaluating the concomitant electrochem. reduction and separation of organic and inorganic model contaminants from water. A highly conductive (40,000 S/m) support free carbon nanotube (CNT) membrane was used as a cathode in a three-electrode electrochem. crossflow membrane filtration system. Diatrizoate (DTZ), a recalcitrant iodinated contrast media; resazurin (RZ), a redox indicator; and hexavalent chromium ion (Cr(VI)), a ‘gold’ standard for characterization of reducing systems, were tested as model contaminants. Voltages of -0.6 to -1.0 V were enough to remove between 90% and 95% of the three model contaminants tested. Removal of Cr(VI) proceeded by reduction to Cr(III) followed by adsorption onto the cathode, which could be efficiently regenerated (up to 95%) applying reversal (anodic) potential. Preliminary findings applying AC current (0.6-1.5 V, 10 Hz-1 kHz) suggest that it could be a feasible approach for detoxification with minimization of membrane clogging. Resazurin was immediately transformed into resorufin to near completion which was in turn further reduced to dihydroresorufin. The reductive transformation of DTZ resulted in almost complete deiodination, leading to the accumulation of 3,5-diacetamidobenzoic acid as the main end-product. Although other five secondary transformation products were detected, four of them were fully deiodinated. Concluding, the proposed electrochem. filtration cell equipped with a highly conductive CNT-cathodic membrane can be regarded as a potential technique for water decontamination and effective dehalogenation of organic compounds

According to the preliminary bioassay studies, all tested mols., especially compounds (I) [R1 = H, R2 = H, R3 = Bn, R4 = CO-C(Me,Me)Et; R1 = 2-Me, R2 = H, R3 = Bn, R4 = CO-C(Me,Me)Et; R1 = 2,4-Cl, R2 = H, R3 = CH2SMe, R4 = CO-i-Pr; R1 = H, R2 = 4-i-Pr-C6H5, R3 = Me, R4 = CO-i-Pr] exhibited significant and broad-spectrum anti-fungal effect in vitro compared to the intermediates M-1, M-2, M-3 and hymexazol. What’s more, the inhibition rate of compounds I (R1 = 2-Me, R2 = H, R3 = Bn, R4 = CO-C(Me,Me)Et; R1 = 4-F, R2 = H, R3 = Bn, R4 = CO-i-Pr; R1 = H, R2 = 4-i-Pr-C6H5, R3 = Me, R4 = CO-i-Pr) against Phomopsis adianticola reached 74.42%, 60.33%, 65.21%, as well as compounds I (R1 = R2 = H, R3 = Bn, R4 = CO-OEt; R1 = H, R2 = 4-i-Pr-C6H5, R3 = Me, R4 = CO-i-Pr) against Sclerotinia sclerotiorum were 65.65%, 74.92%, 61.36%. Further investigation results indicated that compounds I [R1 = H, R2 = H, R3 = Bn, R4 = CO-C(Me,Me)Et; R1 = 2-Me, R2 = H, R3 = Bn, R4 = CO-C(Me,Me)Et; R1 = H, R2 = 4-i-Pr-C6H5, R3 = Me, R4 = CO-i-Pr] presented obviously inhibitory activities against Phomopsis adianticola compared with hymexazol, which might be considered as the basic active framework for further potential fungicides discovery.

The functional expression of olfactory receptors (ORs) is a primary requirement to examine the mol. mechanisms of odorant perception and coding. Functional expression of the rat I7 OR and its trafficking to the plasma membrane was achieved under optimized exptl. conditions in the budding yeast Saccharomyces cerevisiae. The membrane expression of the receptor was shown by Western blotting and immunolocalization methods. Moreover, we took advantage of the functional similarities between signal transduction cascades of G protein-coupled receptor in mammalian cells and the pheromone response pathway in yeast to develop a novel biosensor for odorant screening using luciferase as a functional reporter. Yeasts were engineered to coexpress I7 OR and mammalian Gα subunit, to compensate for the lack of endogenous Gpa1 subunit, so that stimulation of the receptor by its ligands activates a MAP kinase signaling pathway and induces luciferase synthesis. The sensitivity of the bioassay was significantly enhanced using mammalian Golf compared to the Gα15 subunit, resulting in dose-dependent responses of the system. The biosensor was probed with an array of odorants to demonstrate that the yeast-borne I7 OR retains its specificity and selectivity towards ligands. The results are confirmed by functional expression and bioluminescence response of human OR17-40 to its specific ligand, helional. Based on these findings, the bioassay using the luciferase reporter should be amenable to simple, rapid and inexpensive odorant screening of hundreds of ORs to provide insight into olfactory coding mechanisms.

Mammalian olfactory system is the bio-archetype of smell sensor devices. It is based on a very articulated mechanism which translate the odorant capture information performed by the olfactory receptors (ORs) into a code. Finally, the code is sent to the brain for aroma recognition. Our aim is to partially mimic this system to produce a biosensor on nanometric scale. The active part of the device is constituted of nanosomes containing specific ORs. Each nanosome is interfaced with nanoelectrodes and the odorant capture is converted into an elec. signal. Specifically, the elec. response is correlated with the conformational change that a single OR undergoes when it captures a specific odorant mol. An array of nanodevices should be able to produce specific response profiles. In this paper we present a possible theor. framework in which the exptl. results should be embedded.
