The reactivity of 2-benzyl-1,2,3-triazole in palladium-catalyzed direct arylation was studied. The reaction conditions for the selective synthesis of 2-benzyl-4-aryl-1,2,3-triazoles in moderate to high yields using phosphine-free Pd(OAc)2 catalyst and inexpensive KOAc base have been found. Then, from these 4-aryl-1,2,3-triazoles, the palladium-catalyzed C-H bond functionalization of the C5-position allowed the synthesis of the corresponding 4,5-diarylated 2-benzyl-1,2,3-triazoles. This selective 4,5-diarylation of 2-benzyl-1,2,3-triazole was successfully applied for the straightforward building of the π-extended polycyclic heteroaromatic structures phenanthro[9,10-d][1,2,3]triazoles through Pd-catalyzed C4- and C5-intermol. arylations followed by Pd-catalyzed C-H intramol. arylation.

Shi, Xinzhe;Sosa Carrizo, E. Daiann;Cordier, Marie;Roger, Julien;Pirio, Nadine;Hierso, Jean-Cyrille;Fleurat-Lessard, Paul;Soule, Jean-Francois;Doucet, Henri research published 《 C-H Bond Arylation of Pyrazoles at the β-Position: General Conditions and Computational Elucidation for a High Regioselectivity》, the research content is summarized as follows. Direct arylation of most five-membered ring heterocycles are generally easily accessible and strongly favored at the α-position using classical palladium-catalysis. Conversely, regioselective functionalization of such heterocycles at the concurrent β-position remains currently very challenging. Herein, authors report general conditions for regioselective direct arylation at the β-position of pyrazoles, while C-H α-position is free. By using aryl bromides as the aryl source and a judicious choice of solvent, the arylation reaction of variously N-substituted pyrazoles simply proceeds via β-C-H bond functionalization. The β-regioselectivity is promoted by a ligand-free palladium catalyst and a simple base without oxidant or further additive, and tolerates a variety of substituents on the bromoarene. DFT calculations revealed that a protic solvent such as 2-ethoxyethan-1-ol significantly enhances the acidity of the proton at β-position of the pyrazoles and thus favors this direct β-C-H bond arylation. This selective pyrazoles β-C-H bond arylation was successfully applied for the straightforward building of π-extended poly(hetero)aromatic structures via further Pd-catalyzed combined α-C-H intermol. and intramol. C-H bond arylation in an overall highly atom-economical process.

Pan, Shitao;Wang, Xiu;Ni, Chuanfa;Hu, Jinbo research published 《 Nucleophilic trifluoromethylation of azinium salts with Zn(CF3)2·bpy》, the research content is summarized as follows. (Trifluoromethyl)zinc complexes had been widely used in metal-mediated trifluoromethylation reactions. However, direct nucleophilic addition with a (trifluoromethyl)zinc complex was rare. In this article, an unprecedented trifluoromethylation of azinium salts using Zn(CF3)2·bpy as CF3 source, giving 1-(4-methoxybenzyl)-2-(trifluoromethyl)-(1,2-dihydroquinolines/1,2-dihydroisoquinoline) I [R = H, 6-F, 6-Cl, etc.; R1 = CF3, 4-methoxybenzyl; R2 = 4-methoxybenzyl, CF3; Y = N, CH; X = N, CH] as products was described. The latter species were further transformed to 2-trifluoromethylquinolines II [R = H, 6-F, 6-Me, etc.] under oxidative conditions. This work also shows that ligands play an important role in tuning the reactivity of (trifluoromethyl)zinc complexes.

The palladium-catalyzed Suzuki-Miyaura cross-coupling reaction of organohalides and organoborons is a reliable method for carbon-carbon bond formation. This reaction involves a base-mediated transmetalation process, but the presence of a base also promotes competitive protodeborylation. Herein, authors established a Suzuki-Miyaura cross-coupling reaction via Lewis acid-mediated transmetalation of an organopalladium(II) intermediate with organoborons. Exptl. and theor. investigations indicate that the controlled release of the transmetalation-active intermediate enables base-independent transmetalation under heating conditions and enhances the applicable scope of this process. This system enables to avoid the addition of a traditional base and, thus, renders substrates with base-sensitive moieties available. Results from this research further expand the overall utility of cross-coupling chem.

1532-97-4, 4-Bromoisoquinoline, also known as 4-Bromoisoquinoline, is a useful research compound. Its molecular formula is C9H6BrN and its molecular weight is 208.05 g/mol. The purity is usually 95%.

4-Bromoisoquinoline is an aryl halide that can be used in the cross-coupling reaction with other aryl halides. It has been shown to have anticancer activity and to inhibit the growth of tumour cell lines in vitro. This compound is also efficient for inhibiting leukemia cells. 4-Bromoisoquinoline has been shown to have an inhibitory effect on cancer cells through the inhibition of DNA synthesis and RNA transcription, as well as by inducing apoptosis. The mechanism of action may be due to its ability to bind to aromatic hydrocarbons and halides, which leads to thermodynamic changes and vibrational energy transfer., HPLC of Formula: 1532-97-4
