Nguyen, Thao T.;Nguyen, Khang X.;Pham, Phuc H.;Ly, Duc;Nguyen, Duyen K.;Nguyen, Khoa D.;Nguyen, Tung T.;Phan, Nam T. S. research published 《 Copper-catalyzed synthesis of pyrido-fused quinazolinones from 2-aminoarylmethanols and isoquinolines or tetrahydroisoquinolines》, the research content is summarized as follows. Pyrido-fused quinazolinones were synthesized via copper-catalyzed cascade C(sp2)-H amination and annulation of 2-aminoarylmethanols with isoquinolines or pyridines. The transformation proceeded readily in the presence of a com. available CuCl2 catalyst with mol. oxygen as a green oxidant. Moreover, the dehydrogenative cross-coupling of 2-aminoarylmethanols with tetrahydroisoquinolines were explored, in which CuBr exhibited higher catalytic activity than CuCl2. Broad substrate scope with good tolerance of functionalities were observed under the optimized reaction conditions. The bioactive naturally occurring alkaloid rutaecarpine was obtained by this strategy. The remarkable feature of this protocol was that complicated heterocyclic structures were readily achieved in a single synthetic step from easily accessible reactants and catalysts. The pathway to pyrido-fused quinazolinones would be complementary to existing protocols.

Mu, Liying;Fan, Wenjing;Yuan, Xiang-Ai;Huang, Congcong;Li, Dan;Bi, Siwei research published 《 Mechanistic insights into the C(sp3)-H heteroarylation of amides and Fukui function analysis of regioselectivity》, the research content is summarized as follows. A computational study is carried out to understand the mechanism and excellent regioselectivity in metal-free heteroarylation of amides reported by Zhu’s group. The heteroarylation reaction started with the initial generation of key nitrogen-centered radicals via ligand exchange between reactant 1a and initiator PIFA under visible-light irradiation Following, this reaction undergoes four-stages: 1,5-hydrogen atom transfer, C-C coupling, single electron transfer and proton transfer. The C-C coupling step is identified as the selectivity-determining step in which the carbon-centered radical (C) selectively only attacks the carbon atom adjacent to nitrogen of lepidine (2a). And the radical C more easily attacks the protonated 2a, compared with unprotonated 2a, due to significantly lowered SOMO/LUMO energy difference between them to promote this nucleophilic radical addition From the calculated result, we can see that the pos. effect of the acidity of the reaction substrates on the nucleophilic addition to heteroarenes. Fukui functions of different types of heteroarene substrates are calculated to predict the favorable nucleophilic sites. The calculated most favorable reactive sites of heteroarene substrates are well consistent with the exptl. observed ones. This theor. research provides deeper understandings for the underlying mechanism and the origin of exclusive regioselectivity of the heteroarylation of amides.

Momiyama, Norie;Izumiseki, Atsuto;Ohtsuka, Naoya;Suzuki, Toshiyasu research published 《 Correlations between Substituent Effects and Catalytic Activities: A Quantitative Approach for the Development of Halogen-Bonding-Driven Anion-Binding Catalysts》, the research content is summarized as follows. A quant. approach for the development of halogen-bonding-driven anion-binding catalysts was studied using 4-substituted perfluorinated iodobenzene. 19F NMR titrations were used to determine the binding constants K for chloride, and their catalytic activities were evaluated in the allylation reaction of a N-activated pyridine. We discovered that the log K and product yields were linearly correlated, and that they were dependent on the Hammett substituent parameter, σmeta (r2=0.99). This linear correlation provided a quant. predictive model for both the binding constant and the reaction yield. Concomitantly, this efficiently permitted the development of a highly active anion-binding catalyst, namely 4-CNC6F4I (K=489±5 M-1). Addnl., the catalytic activity of 4-CNC6F4I was established in the allylation and crotylation of N-activated isoquinolines (7 examples). Overall, this approach highlights the value of quant. anal. by exploring exptl. informed correlations in the development of halogen bond donor catalysts.

Mahmoodi, Niusha;Harijan, Rajesh K.;Schramm, Vern L. research published 《 Transition-State Analogues of Phenylethanolamine N-Methyltransferase》, the research content is summarized as follows. Phenylethanolamine N-methyltransferase (PNMT) is a critical enzyme in catecholamine synthesis. It transfers the Me group of S-adenosylmethionine (SAM) to catalyze the synthesis of epinephrine from norepinephrine. Epinephrine has been associated with diverse human processes, including the regulation of blood pressure and respiration, as well as neurodegeneration found in Alzheimer’s disease. Human PNMT (hPNMT) proceeds through an SN2 transition state (TS) in which the transfer of the Me group is rate limiting. TS analog enzyme inhibitors are specific for their target and bind orders of magnitude more tightly than their substrates. Mols. resembling the TS of hPNMT were designed, synthesized, and kinetically characterized. This new inhibitory scaffold was designed to mimic the geometry and electronic properties of the hPNMT TS. Synthetic efforts resulted in a tight-binding inhibitor I with a Ki value of 12.0 nM. This is among the first of the TS analog inhibitors of methyltransferase enzymes to show an affinity in the nanomolar range. Isothermal titration calorimetry (ITC) measurements indicated neg. cooperative binding of inhibitor to the dimeric protein, driven by favorable entropic contributions. Structural anal. revealed that I binds to hPNMT by filling the catalytic binding pockets for the cofactor (SAM) and the substrate (norepinephrine) binding sites.

Lima, Fabio;Andre, Jerome;Marziale, Alexander;Greb, Andreas;Glowienke, Susanne;Meisenbach, Mark;Schenkel, Berthold;Martin, Benjamin;Sedelmeier, Joerg research published 《 Continuous Flow as Enabling Technology: Synthesis of Heteroaromatic Sulfinates as Bench Stable Cross-Coupling Partners》, the research content is summarized as follows. An enabling continuous flow setup for handling of unstable organolithium intermediates and synthesis of heteroaryl sulfinates on a multigram scale is described. The developed continuous flow process allows for the synthesis and simple isolation of heteroaryl sulfinates which are otherwise challenging to access in classical batch mode. The lithium sulfinate salts prepared by this method were shown to be efficient reaction partners in palladium catalyzed C(sp2)-C(sp2) cross-coupling to access medicinally relevant bis-heteroaryl motifs.

Category: isoquinoline, 4-Bromoisoquinoline, also known as 4-Bromoisoquinoline, is a useful research compound. Its molecular formula is C9H6BrN and its molecular weight is 208.05 g/mol. The purity is usually 95%.

4-Bromoisoquinoline is an aryl halide that can be used in the cross-coupling reaction with other aryl halides. It has been shown to have anticancer activity and to inhibit the growth of tumour cell lines in vitro. This compound is also efficient for inhibiting leukemia cells. 4-Bromoisoquinoline has been shown to have an inhibitory effect on cancer cells through the inhibition of DNA synthesis and RNA transcription, as well as by inducing apoptosis. The mechanism of action may be due to its ability to bind to aromatic hydrocarbons and halides, which leads to thermodynamic changes and vibrational energy transfer., 1532-97-4.
