Structural alerts are widely accepted in chem. toxicol. and regulatory decision support as a simple, transparent means to flag potential chem. hazards or group compounds into categories for read-across. However, there is a growing concern that alerts disproportionally flag chems. as toxic, which questions their reliability as toxicity markers. Conversely, rigorously developed, properly validated statistical QSAR models can accurately, reliably predict the toxicity of a chem.; however, their use in regulatory toxicol. has been hampered by a lack of transparency and interpretability. This work demonstrated that contrary to the common perception of QSAR models as black boxes, they can identify statistically significant chem. substructures (QSAR-based alerts) which influence toxicity. The authors show using case studies, that the mere presence of structural alerts in a chem., irresp. of the derivation method (expert- or QSAR-based), should be perceived only as hypotheses of possible toxicol. effect. A new approach is proposed which synergistically integrates structural alerts and rigorously validated QSAR models for a more transparent, accurate safety assessment of new chems.

We test the Kozameh-Quiroga formalism evolving the equations of motion for the center of mass, the intrinsic angular momentum, and several other global variables, for black hole binary coalescence. First, we evolve the equations of motion using 777 simulations from the RIT catalog of numerical data of ψ4 [J. Healy and C. O. Lousto, Third RIT binary black hole simulations catalog, Phys. Rev. Chinese horse bean-chili-paste (CHCP), a fermented condiment in China, is traditionally manufactured through naturally spontaneous semi-solid fermentation procedures without intentionally inoculated microorganisms. The aim of this study was to investigate the effect on microbiota and quality variations during CHCP fermentation by inoculation of selected autochthonous microorganisms Bacillus amyloliquefaciens and Candida versatilis. The results showed that relative abundance of Bacillus in the samples inoculated with B. amyloliquefaciens were increased from about 0.6% to almost 25%, and the batches bio-augmented with C. versatilis exhibited clearly 0.7 Lg copies/g higher biomass than that of the other samples. By bio-augmentation, six enzyme activities, namely acid protease, leucine aminopeptidase, α-amylase, cellulose, β-glucosidase and esterase, were considerably enhanced. As a result, inoculation of these two strains exhibited significant effect on the volatile profiles of CHCP. B. amyloliquefaciens herein was found to contribute mainly to the accumulation of acids, sulfur-containing compounds and pyrazines, whereas C. versatilis was considerably associated with the formation of alcs., esters and phenols. This study proved that combination of B. amyloliquefaciens and C. versatilis could obtain more extensive aroma profiles, especially for the enrichment of miso-like and fruity flavors, which could provide a guideline for the tailored control of CHCP fermentation process.

Alkalization is a process to improve color, dispersibility and flavor of cocoa powder but is likely to have a neg. effect on the phytochems. Hereto, the impact of alkalization degree (none, medium and high) on the potential mood-enhancing compounds corresponding to the four levels of the mood pyramid model (flavanols, methylxanthines, biogenic amines and orosensory properties) was investigated. The phytochem. content, analyzed via UPLC-HRMS, showed reduction of specific potential mood-enhancing compounds upon alkalization, implying a decrease in bitterness and astringency. Moreover, volatile compounds anal. via HS-SPME-GC-MS indicated that alkalization reduced the levels of volatile compounds, responsible for acidity, fruity, floral and cocoa aromas. With respect to the orosensory properties, the cocoa powder palatability was suggested to be increased due to reduced acidity, bitterness, and astringency, while the desired volatile compounds were reduced. However, sensorial anal. is required to link the volatile results with the overall effect on the flavor perception.

A total of 58 volatile components were found in natural-type douchi compared to artificial-type with 42 identified. Two cocoa types are predominantly being cultivated in Ecuador, the highly appreciated fine Nacional and the high-yielding and disease-resistant CCN51, classified as bulk. With the introduction of foreign germplasm in Ecuador and the international demand for high-quality Nacional cocoa, the present study aims to characterize and compare the flavor profile of Nacional hybrids (EET103, EET559, EET576 and EET577) and CCN51, and to assess the effect of the genotype. The fermented dried beans were processed into liquor and their sensory properties, volatile composition and odor fingerprint were analyzed by trained panelists, headspace-solid phase microextraction gas chromatog.-mass spectrometry (HS-SPME-GC-MS), and GC electronic nose (e-nose), resp., combined with multivariate statistics. Seventy volatiles were identified and semi-quantified, among them, 16 and 14 volatiles conferring fruity and floral aroma notes, resp. CCN51 was mainly characterized by undesirable odor-active volatiles, whereas the EET cultivars contained the highest concentration of most fruity and floral volatiles, with differences in between EET cultivars. Sensory data highlighted organoleptic differences between the fine and bulk liquors, i.e. the fruity and acid EET flavor strongly differed from the bitter, astringent and off-flavor attributes of CCN51. Principal component anal. (PCA) discriminated firstly between fine and bulk, and secondly between EET identity. Partial least squares anal. identified possible correlations between sensory attributes and volatile compounds Besides, e-nose proved its usefulness for rapid assessment of the liquors’ sensory profile. The results provide a foundation for understanding the genotype’s contribution to and the compositional basis of Ecuadorian fine and bulk cocoa flavor.

The quality of 10 buckwheat honeys, collected from Italian and east European beekeepers declaring to produce monofloral honey, were evaluated by means of their pollen, physicochem., phenolic and volatile composition data. The results of the traditional analyses and in particular elec. conductivity, optical rotation, pH and sugar composition revealed some poorly pure samples that could not fit in the buckwheat tipol. Honey volatiles, analyzed by solid phase microextraction (SPME) and gas chromatog.-mass-spectrometry (GC/MS), showed more than 100 volatile compounds, most of them present in all honey samples but with quant. variation. Besides many furfural derivates, 3-methylbutanoic acid was the main volatile compound found in most of honeys. Also the presence of 2- and 3-methylbutanal and phenylacetaldehyde confirmed the typical buckwheat aroma of some studied samples, corroborating physicochem. data. The HPLC phenolic profile was similar across the samples and p-hydroxybenzoic and p-coumaric acids proved to be the main components.

Since March 2013, it is no longer possible to market in the European Union (EU) cosmetics containing new ingredients tested on animals. Although several in silico alternatives are available and achievements have been made in the development and regulatory adoption of skin sensitization non-animal tests, there is not yet a generally accepted approach for skin sensitization assessment that would fully substitute the need for animal testing. The aim of this work was to build a defined approach (i.e. a predictive model based on readouts from various information sources that uses a fixed procedure for generating a prediction) for skin sensitization hazard prediction (sensitizer/non-sensitizer) using Local Lymph Node Assay (LLNA) results as reference classifications. To derive the model, we built a dataset with high quality data from in chemico (DPRA) and in vitro (KeratinoSens and h-CLAT) methods, and it was complemented with predictions from several software packages. The modeling exercise showed that skin sensitization hazard was better predicted by classification trees based on in silico predictions. The defined approach consists of a consensus of two classification trees that are based on descriptors that account for protein reactivity and structural features. The model showed an accuracy of 0.93, sensitivity of 0.98, and specificity of 0.85 for 269 chems. In addition, the defined approach provides a measure of confidence associated to the prediction.
