An improved route for the synthesis of N,N’-diindolyl methane (DIM) glycosides has been developed by using Fe/Al pillared clay catalyst. In-silico pharmacokinetics followed by in-vitro studies like aqueous solubility, lipophilicity, P-glycoprotein (P-gp) dependent ATPase activity, permeability, plasma protein binding, RBC partitioning, metabolic stability in different liver microsomes and its in-vitro-in-vivo extrapolation were conducted for the most potent derivative namely NGD16. The compound was found to have low solubility, optimum lipophilicity, no P-gp inhibitory activity, intermediate permeability, high plasma protein binding, low RBC partitioning, acceptable metabolic stability in rat liver microsomes (RLM) as well as human liver microsomes (HLM) with intermediate hepatic extraction ratio.

The human olfactory system can recognize and discriminate a large number of different odorant mols. The detection of chem. distinct odorants starts with the binding of an odorant ligand to a specific receptor protein on the olfactory neuron cell surface. To address the problem of olfactory perception at a mol. level, we have expressed and characterized different olfactory receptors with several expression systems. Here we provide the first documentation of functional expression of odorant receptors using the Semliki Forest virus system. The human odorant OR 17-40 receptor and the rat 17 receptor were functionally expressed in vertebrate kidney cells (HEK293) using recombinant Semliki Forest viruses. Receptors were expressed as a fusion protein with the N-terminal membrane import sequence of the guinea pig serotonin receptor. Experiments employing the Ca2+-sensitive dye fura-2 revealed a fast, transient increase in the [Ca2+]i after application of the specific agonists helional and octanal to HEK293 cells infected with viruses containing RNA for the human odorant OR 17-40 receptor and the rat 17 receptor, resp.

Regarding the chemotaxis of sperms, new insights have been gained during the last 20 years. Olfactory receptors are expressed on the flagellar midpiece of human spermatogenic cells. One of them, OR1D2, is also expressed in the olfactory epithelium. This receptor has been suggested to play a role in sperm chemotaxis and thus in fertility. As OR1D2 is activated by bourgeonal, the aim of the study was to investigate whether patients with idiopathic infertility would exhibit a decreased olfactory sensitivity toward bourgeonal. Participants were 14 patients with idiopathic infertility and 23 controls (all young fathers). After having ascertained normosmia, odor thresholds and intensity ratings for the pleasant and flowery odors of bourgeonal, helional, and phenylethylalc. were obtained. As a result, patients had specifically decreased intensity ratings for bourgeonal. It suggests that men with unexplained infertility tend to be less suprathreshold sensitive toward the odor of bourgeonal but not to that of other floral odors. It may be speculated that the decreased olfactory sensitivity relates to a decreased functionality of OR1D2, which in turn may be linked to idiopathic infertility.

The European Food Safety Authority highlights the beneficial effects of olive oil phenols, mainly, secoiridoids. Nevertheless, the metabolism of secoiridoids in humans has not been fully elucidated. This research evaluated the metabolism of secoiridoids in humans after intake of olive oils with diverse phenolic profiles. For this purpose, three extra virgin olive oils (EVOOs) were ingested by six volunteers at scheduled meals, and urine samples were collected the following morning for subsequent LC-MS/MS anal. Using untargeted anal., urinary metabolites revealed representative patterns associated with the various olive oil phenolic contents in absolute and relative terms. We were able to identify metabolites obtained through phase I, phase II, and microbial metabolism with discrimination between tyrosol and hydroxytyrosol derivatives Metabolism of phenols is differentially activated as a function of the olive oil secoiridoids content, and this proof-of-concept study shows how urinary metabolites represent olive oil phenolic content.

2-(P-Ph substituted styryl)-furans were synthesized and studied the excited state and anti oxidation properties using absorption, fluorescence, d. functional theory and DPPH radical scavenging assay. 2-(P-hydroxy Ph styryl)-furan (6) exhibits good antioxidant properties with IC50 âˆ?40μM as compared to the amine, chloro, cyano methoxy and nitro substituted styryl compounds (1-5, 7-8) (IC50 > 150μM). It is shown that furans 1-3 with strong electron withdrawing nitro, cyano and chloro substituent do not show antioxidant properties. The O-H bond dissociation energy (BDE) of p-hydroxy substituted furan compound is comparable to the BDE of vitamin E. The antioxidant activity of furan compound is further reduced with increasing the dipolar nature of the mol. From the thermodn. parameters and antioxidant properties, it is shown that there is a correlation of antioxidant properties with the ground state dipole moment, X-H bond dissociation energy (X: O; N) and proton affinity (PA). In presence of the furan compound, the quenching of DPPH radical is occurred predominantly through hydrogen atom transfer mechanism.

In this study, the influence of the duration and temperature of the roasting process on the formation of these malt characteristics was monitored. The results indicated that the development of caramel-like flavor components was highly influenced by the roasting temperature and moisture content of the malt kernels, and the formation of pyrazines strongly increased at 180°C. In addition, the evolution of malt characteristics such as color and reducing power was greatly influenced by the applied processing parameters. Finally, the results indicated flavor development was not coupled with color formation.

This procedure offers several advantages such as: very easy reaction conditions, simple work-up, or purification, excellent yields, high purity of the desired product, atom economy, and short reaction times. The superparamagnetic nanocatalyst is magnetically separable and kept stabile after recycling for at least five consecutive runs without detectable activity loss.

Malt is usually expected to be stable during 12 mo of storage. However, in practice many brewers notice changes in malt aroma during storage. The oxidative stabilities of pilsner malt and roasted malt were evaluated during a 12 mo storage at different temperatures (10 and 20 °C) and water activities (0.231 and 0.432). The radical content in malt kernels was measured by ESR spectroscopy and the volatile profile of the resulting sweet worts was measured by head-space anal. followed by GC-MS anal. The storage of malt resulted in oxidative reactions and a large change of the volatile profile of the resulting worts. Roasted malt was much more unstable than pilsner malt, as illustrated by a higher initial radical intensity, larger radical decay during storage and a larger change in the volatile profile of the wort with increased amounts of lipid oxidation products. For both roasted malt and pilsner malt, good correlations were found between radical decay and changes in the volatile profile of the wort, where high temperature and high water activity resulted in the largest changes. During the 12 mo of storage, the sugar extract of the wort made from the malts remained constant and was not affected by the chem. changes. This study suggests that chem. changes occurring in malts during less than 12 mo of storage may potentially affect the aroma of beer, and that water activity and storage temperature should both be kept low in order to maintain a high malt quality. Dahuang zhechong pill (DHZCP) is a famous traditional Chinese medicine prescription, which is widely used in the treatment of liver diseases. However, due to the lack of a dynamic DHZCP profile, the in vivo pharmacokinetics of active ingredients within this medicine remains unknown. In this paper, a rapid, sensitive and reliable UHPLC-MS/MS method was used to determine the content of 19 characteristic constituents of DHZCP in rat plasma, including rhein, emodin, chrysophanol, physcion, aloeemodin, p-methoxyphenylacetic acid, hypoxanthine nucleoside, wogonin, wogonoside, baicalin, norwogonin, naringenin, nutmeg acid, paeoniflorin, verbascoside, rhodiola glucoside, forsythoside A, formononetin, and glycyrrhizic acid. An Agilent Extend-C18 column (2.1 mm x 100 mm, 1.8 μm) was used to sep. the 19 characteristic constituents, with a mobile phrase of (A) 0.1% formic acid and (B) acetonitrile. The constituents were detected in neg. ion mode with multiple reactions monitoring (MRM). The established UHPLC-MS/MS method had good linearity, with a coefficient of determination (r2) of >0.99. The daytime and intra-day precision were less than 12%, and the accuracy ranged from -9.56% to 7.82%. The stability, extraction recovery, and matrix effect met the requirements. The method was successfully applied to the pharmacokinetic study of these nineteen characteristic constituents after oral administration of DHZCP. UHPLC-MS/MS was used for the first time to study the pharmacokinetics of the characteristic chem. constituents in DHZCP, which provided reference and theor. guidance for further clarification of its pharmacodynamic basis.
