Fused thieno[3,2-d]thiazoles were synthesized via a coupling of acetophenone ketoximes, arylacetic acids, and elemental sulfur in the presence of Li2CO3 base. Functionalities including chloro, bromo, fluoro, trifluoromethyl, and pyridyl groups were compatible with reaction conditions. High yields and excellent regioselectivities were obtained even if meta-substituted ketoxime acetates were used. Et esters of heteroarylacetic acids were competent substrates, which is very rare in the literature. This method would offer a convenient protocol to afford polyheterocyclic structures from simple substrates.

Based on two sterically distinct cyclometalating ligands, the nonracemic rhodium(III) catalyst could conveniently be accessed in an enantiomerically pure fashion (>99% ee) via an established auxiliary-mediated approach. Comparison of the catalytic performance with the related C2-sym. rhodium catalysts revealed the explicit superiority of the non-C2-sym. design for the presented α-halogenation reactions, which were generally featured by a very simple synthetic protocol.

Conformational anal. of new para-substituted 2-bromo-2-Pr 2-aryl-acetates (Y = H, OMe, Cl, and NO2) (R1), ibuprofen (R2), and naproxen (R3) analogs using IR (IR) spectroscopy and theor. calculations was performed to determine the preferential conformers of these compounds in solvents with increasing polarity (CCl4, CH3Cl, and CH3CN). The aryl-bromo-esters were synthesized via the esterification of 2-bromo-2-methylpropan-1-ol and the corresponding carboxylic acids, with good yields (∼36-70%). The IR spectra showed that these compounds presented only one conformation, and the exptl. data were supported by the theor. results obtained by d. functional theory (DFT) calculations using the 6311+G (2df, 2p) basis set. The calculations revealed that all the studied compounds presented two stable geometric conformations, which agrees with the data obtained exptl. in CCl4. Theses conformers are stabilized by intramol. hydrogen bonds. However, the orbital interaction calculations using the natural bond orbital (NBO) method showed that the ηO→σ*C-C,ηO→σ*C-O, ηO→σ*C-O and ηO→π*C-O hyper-conjugations are the main interactions that stabilize the conformations. The compounds preferentially adopt the anti-conformation because the steric effect between the gauche bromo and oxygen atoms overrides the hyper-conjugative interactions, in addition to the stabilizing σC-H→ σ*C-Br interactions in the conformers.

In this paper, a radical tandem C-N coupling strategy to efficiently construct aromatic tertiary amines from com. available carboxylic acids and nitroarenes was developed. The use of two different carboxylic acids also allowed for the concise synthesis of nonsym. aromatic tertiary amines in satisfactory yields. Mechanistic studies suggested the intermediacy of the arylamine-(TPP)Fe(III) species and might provide a possible evidence for an SH2 (bimol. homolytic substitution) pathway in the critical C-N bond formation step.

Hantzsch ester (HE) functions as a cheap, green and strong photoreductant to facilitate radical generation and also engages in the Ni-catalytic cycle to restore the reactive species. With this dual role, HE allows for the coupling of a large variety of radicals (1°,2°, benzylic, α-oxy & α-amino) with aroyl and alkanoyl moieties, a new feature in reactions of this type. With both precursors deriving from abundant carboxylic acids, this protocol is a welcome addition to the organic chem. toolbox. The reaction proceeds under mild conditions without the need for toxic metal reagents or bases and shows a wide scope, including pharmaceuticals and complex mol. architectures.

Herein, a copper-mediated, net-oxidative decarboxylative coupling of carboxylic acids with diverse nucleophiles under visible-light irradn is reported. Preliminary mechanistic studies suggest that the relevant chromophore in this reaction is a Cu(II) carboxylate species assembled in situ. Authors propose that visible-light excitation to a ligand-to-metal charge transfer (LMCT) state results in a radical decarboxylation process that initiates the oxidative cross-coupling. The reaction is applicable to a wide variety of coupling partners, including complex drug mols., suggesting that this strategy for cross-nucleophile coupling would facilitate rapid compound library synthesis for the discovery of new pharmaceutical agents.
