Maleimide chem. is widely used in the site-selective modification of proteins. However, hydrolysis of the resultant thiosuccinimides is required to provide robust stability to the bioconjugates. Herein, we present an alternative approach that affords simultaneous stabilization and dual functionalisation in a one pot fashion. By consecutive conjugation of a thiol and an amine to dibromomaleimides, we show that aminothiomaleimides can be generated extremely efficiently. Furthermore, the amine serves to deactivate the electrophilicity of the maleimide, precluding further reactivity and hence generating stable conjugates. We have applied this conjugation strategy to peptides and proteins to generate stabilized trifunctional conjugates. We propose that this stabilization-dual modification strategy could have widespread use in the generation of diverse conjugates.

Though d. functional theory is widely accepted as one of the most successful developments in theor. chem. in the past few decades, the knowledge of how to apply this new electronic structure theory, to help us better understand chem. processes and transformations, is still an unaccomplished task. The information-theor. approach is emerging as a viable option for that purpose in the recent literature, providing new insights about steric effect, cooperativity, electrophilicity, nucleophilicity, stereoselectivity, homochirality, etc. In this work, based on the result from a recent paper by one of the authors [J. Chem. Phys., 2019, 151, 141103], the authors present two quantifications of the relative Fisher information and discuss their physiochem. properties and possible applications. To that end, their anal. properties have been elucidated. They have also been applied to six categories of systems to illustrate their applicability. A better descriptor to quantify the single bond rotation barrier has been obtained. The relative Fisher information can also simultaneously determine electrophilicity and nucleophilicity, and effectively describe helical structures with different homochiral and heterochiral propensities. As integral parts of the information-theor. approach, these newly introduced quantities will provide us with more anal. tools toward the long-term goal of crafting a chem. reactivity theory in the d.-based language.

The synthesis of highly acidic metal-organic frameworks (MOFs) has attracted significant research interest in recent years. We report here the design of a strongly Lewis acidic MOF, ZrOTf-BTC, through two-step transformation of MOF-808 (Zr-BTC) secondary building units (SBUs). Zr-BTC was first treated with 1 M hydrochloric acid solution to afford ZrOH-BTC by replacing each bridging formate group with a pair of hydroxide and water groups. The resultant ZrOH-BTC was further treated with trimethylsilyl triflate (Me3SiOTf) to afford ZrOTf-BTC by taking advantage of the oxophilicity of the Me3Si group. ESR spectra of Zr-bound superoxide and fluorescence spectra of Zr-bound N-methylacridone provided a quant. measurement of Lewis acidity of ZrOTf-BTC with an energy splitting (ΔE) of 0.99 eV between the πx* and πy* orbitals, which is competitive to the homogeneous benchmark Sc(OTf)3. ZrOTf-BTC was shown to be a highly active solid Lewis acid catalyst for a broad range of important organic transformations under mild conditions, including Diels-Alder reaction, epoxide ring-opening reaction, Friedel-Crafts acylation, and alkene hydroalkoxylation reaction. The MOF catalyst outperformed Sc(OTf)3 in terms of both catalytic activity and catalyst lifetime. Moreover, we developed a ZrOTf-BTC@SiO2 composite as an efficient solid Lewis acid catalyst for continuous flow catalysis. The Zr centers in ZrOTf-BTC@SiO2 feature identical coordination environment to ZrOTf-BTC based on spectroscopic evidence. ZrOTf-BTC@SiO2 displayed exceptionally high turnover numbers (TONs) of 1700 for Diels-Alder reaction, 2700 for epoxide ring-opening reaction, and 326 for Friedel-Crafts acylation under flow conditions. We have thus created strongly Lewis acidic sites in MOFs via triflation and constructed the MOF@SiO2 composite for continuous flow catalysis of important organic transformations.

Importantly, the key binuclear cyclopalladium intermediate bridged by acetate anion was unambiguously characterized by X-ray diffraction, which solidly supports the proposed mechanism. The practical utility was further depicted as facile access to oral antiplatelet agent Clopidogrel racemate via subsequent two-step derivatization.

Photoinduced electron transfer (PET) is a phenomenon whereby the absorption of light by a chem. species provides an energetic driving force for an electron-transfer reaction1-4. This mechanism is relevant in many areas of chem., including the study of natural and artificial photosynthesis, photovoltaics and photosensitive materials. In recent years, research in the area of photoredox catalysis has enabled the use of PET for the catalytic generation of both neutral and charged organic free-radical species. These technologies have enabled previously inaccessible chem. transformations and have been widely used in both academic and industrial settings. Such reactions are often catalyzed by visible-light-absorbing organic mols. or transition-metal complexes of ruthenium, iridium, chromium or copper5,6. Although various closed-shell organic mols. have been shown to behave as competent electron-transfer catalysts in photoredox reactions, there are only limited reports of PET reactions involving neutral organic radicals as excited-state donors or acceptors. This is unsurprising because the lifetimes of doublet excited states of neutral organic radicals are typically several orders of magnitude shorter than the singlet lifetimes of known transition-metal photoredox catalysts7-11. Here we document the discovery, characterization and reactivity of a neutral acridine radical with a maximum excited-state oxidation potential of -3.36 V vs. a SCE, which is similarly reducing to elemental lithium, making this radical one of the most potent chem. reductants reported12. Spectroscopic, computational and chem. studies indicate that the formation of a twisted intramol. charge-transfer species enables the population of higher-energy doublet excited states, leading to the observed potent photoreducing behavior. We demonstrate that this catalytically generated PET catalyst facilitates several chem. reactions that typically require alkali metal reductants and can be used in other organic transformations that require dissolving metal reductants.

We synthesized two kinds of monocarbamothioates TZO(R) containing a thiazolidin-2-one (TZO) skeleton and TZD(R) containing a thiazolidine-2,4-dione (TZD) skeleton. In the reaction of TZO(R) and phenoxypropyrenesulfide (PPS) using tetrabutylammonium chloride (TBAC) as a catalyst in 1-methyl-2-pyrrolidinone at 60°C, anionic ring-opening polymerization of PPS proceeded to give the corresponding linear polysulfides, polyPPS, with Mn = 75 860-206 700 (Mw/Mn = 1.54-1.73) in high yield. In contrast, in the reaction of TZD(R) and PPS, the continuous insertion reaction of PPS into TZD(R), i.e., living ring-expansion polymerization, occurred to afford cyclic polysulfides TZD(R)-c-poly(PPS)n with Mn = 1870-33 420 (Mw/Mn = 1.07-1.26) in high yield. The ring size could be controlled by the feed ratio of TZD(R)/PPS in the range of 1/5-1/100.
