A new series of 2-(5-(substituted-benzyl)-1,3,4-thiadiazol-2-yl)-substituted-isoindoline-1,3-dione I [R = H, 2-Me, 4-Cl, 4-OMe, 2,4-di-Cl; R1 = H, 4-Me, 3-NO2] was synthesized by the reaction of 2-amino-5-(substituted)-benzyl-1,3,4-thiadiazole with substituted phthalic anhydride under solvent free conditions and the structures of the compounds were confirmed by IR and NMR. Representative compounds were screened for their anti-microbial activity against Gram-neg. bacteria, E. coli and P. aeruginosa and Gram-pos. bacteria, S. aureus, and C. diphtheriae using disk diffusion method. Some of these compounds were found to exhibit excellent antibacterial activity.

In this study, deep eutectic solvents (DESs) were used as green and eco-friendly media for the synthesis of substituted 2-mercaptoquinazolin-4(3H)-ones I [R1 = H, 6-I, 6-Br, 7-Cl, 6,8-dichloro; R2 = Me, Ph, Bn, etc.] from different anthranilic acids and aliphatic or aromatic isothiocyanates. A model reaction on anthranilic acid and Ph isothiocyanate was performed in 20 choline chloride-based DESs at 80 °C to find the best solvent. Based on the product yield, choline chloride:urea (1:2) DES was found to be the most effective, while DESs acted both as solvents and catalysts. Desired compounds were prepared with moderate to good yields using stirring, microwave-assisted, and ultrasound-assisted synthesis. Significantly higher yields were obtained with mixing and ultrasonication (16-76%), while microwave-induced synthesis showed lower effectiveness (13-49%). The structures of the synthesized compounds were confirmed by 1H and 13C NMR spectroscopy.

A gallium-catalyzed C-H propargylation of electron-rich arenes using bromoallenes is described. The development of this reaction was first hampered by a side hydroarylation process catalyzed by adventitious protons, easily generated from the solvent (1,2-dichloroethane). This hidden Bronsted acid catalysis could be bypassed by using K2CO3 as insoluble base. The unexpected compatibility of this base with the Lewis acid catalyst GaCl3 allowed a strictly gallium-catalyzed process that was thoroughly studied by kinetics, 71Ga NMR experiments and DFT computations. The GaCl3/K2CO3 system triggers the C-H propargylation of a wide range of arenes and heteroaromatics using a variety of bromoallenes.

The reaction occurred through an acid-promoted Meinwald rearrangement, producing arylacetyl cyanides, followed by an addition-elimination process with nitrogen or oxygen-containing nucleophilic amines, alcs. or water. The current method provided a new application of the tandem Meinwald rearrangement.
