After stirring for 3 hours, ether (400 mL) is added, followed by addition of hexanes (1 L). Solution stirred overnight and conc in vacuo, ether (200 mL) and hexanes (400 mL) is added, stirred overnight. The ppt is filtered, air dried and mixed with 20 g of POS and toluene (150 mL). The solution is heated to reflux for 3 h, neutralized with NAHCO3. Extracted the solution with CH2CI2. Organic layer then dried with sodium sulfate and conc in vacuo, yield 8 g (72%) of 1, 8-Dichloro-isoquinoline. MS: 198

Portions(65 mg) of resins 15 were placed in reactor wells (12 mL) of anautomated synthesizer reaction block (40-well format) (AdvancedChemTech). To each well was added a solution of appropriate carboxylicacid (see Tables 1?3) (0.3 M), PyBOP (0.3 M) and HOBt 6-Cl(0.3 M) and DIPEA (0.6 M) in dry DMF (2 mL). The suspensionswere vortexed at 300 rpm over a period of 5 h under an argonatmosphere. The crude product was purified by flash column chromatography; (silica; EtOAc in Heptane, 0/100 to 35/65). The desired fractions were collected and concentrated in vacuo. The residue was dissolved in DCM, HC1 (5 N in 2-propanol) was added and the mixture was concentrated in vacuo. The mixture was stirred at room temperature for 0.5 hours. The reaction mixture was diluted by adding chloroform, and washed with a saturated sodium bicarbonate aqueous solution and saturated brine. The organic layer was dried over anhydrous magnesium sulfate, and concentrated under reduced pressure. The obtained residue was dissolved in acetic anhydride (78.0 mL), and stirred for 1 hour under reflux. A 2.0 mol/L sodium hydroxide aqueous solution (156 mL) was added to the residue obtained by concentrating the reaction mixture under reduced pressure, and the mixture was stirred for 2 hours under reflux. The reaction mixture was cooled to room temperature, and neutralized with a 2.0 mol/L hydrochloric acid aqueous solution.
