Chen, Xiang published the artcileDirect transfer of tri- and di-fluoroethanol units enabled by radical activation of organosilicon reagents, HPLC of Formula: 5961-59-1, the main research area is trifluoromethyl difluoromethyl alc preparation; organosilicon reagent radical activation allylation alkylation alkenylation.

The two organosilicon reagents (such as., 1-((dimethyl)(phenyl)silyl)-2,2,2-trifluoroethan-1-ol and 1-((phenyl)(dimethyl)silyl)-2,2-difluoroethanol) for the transfer of trifluoroethanol and difluoroethanol units into mols. have been described. Through intramol. C-Si bond activation by alkoxyl radicals, these reagents were applied in allylation, alkylation and alkenylation reactions, enabling efficient synthesis of various tri(di)fluoromethyl group substituted alcs. e.g., I and, II. The broad applicability and general utility of the approach are highlighted by late-stage introduction of these fluoroalkyl groups to complex mols., and the synthesis of antitumor agent Z and its difluoromethyl analog Z’ III (R = CF3, CF2H).

Bavan, Selvan published the artcileDiscovery of novel ligands for mouse olfactory receptor MOR42-3 using an in silico screening approach and in vitro validation, Name: 5-Methyl-2-phenylhex-2-enal, the main research area is Xenopus oocyte olfactory receptor heterologous odorant.

The ligands for many olfactory receptors remain largely unknown despite successful heterologous expression of these receptors. Understanding the mol. receptive range of olfactory receptors and deciphering the olfactory recognition code are hampered by the huge number of odorants and large number of olfactory receptors, as well as the complexity of their combinatorial coding. Here, we present an in silico screening approach to find addnl. ligands for a mouse olfactory receptor that allows improved definition of its mol. receptive range. A virtual library of 574 odorants was screened against a mouse olfactory receptor MOR42-3. We selected the top 20 candidate ligands using two different scoring functions. These 40 odorant candidate ligands were then tested in vitro using the Xenopus oocyte heterologous expression system and two-electrode voltage clamp electrophysiol. We exptl. confirmed 22 of these ligands. The candidate ligands were screened for both agonist and antagonist activity. In summary, we validated 19 agonists and 3 antagonists. Two of the newly identified antagonists were of low potency. Several previously known ligands (mono- and dic-arboxylic acids) are also confirmed in this study. However, some of the newly identified ligands were structurally dissimilar compounds with various functional groups belonging to aldehydes, phenyls, alkenes, esters and ethers. The high pos. predictive value of our in silico approach is promising. We believe that this approach can be used for initial deorphanization of olfactory receptors as well as for future comprehensive studies of mol. receptive range of olfactory receptors.

Wols, B. A. published the artcilePredicting pharmaceutical degradation by UV (MP)/H2O2 processes: A kinetic model, Recommanded Product: Diatrizoic Acid, the main research area is pharmaceutical photolysis water wastewater UV hydrogen peroxide kinetic model.

A kinetic model for the degradation of organic micropollutants (OMPs) by MP UV lamps was developed. The model includes (photo)chem. reactions of H2O2 and nitrate; reactions with the most important water matrix components; changes in pH; and transient radical concentrations Three pathways of organic micropollutant degradation are involved: direct photolysis, hydroxyl radical reactions and carbonate radical reactions. The model was validated using collimated beam experiments for a group of 35 pharmaceuticals in synthetic and natural water matrixes. For MP UV with 10 mg/L H2O2, good agreement between the modeled and measured degradation rates was found. For MP UV without H2O2, most of the pharmaceuticals were well predicted, except for the sulfonamides. A sensitivity anal. on different water matrix constituents was performed for MP UV with H2O2, NO3- and both H2O2 and NO3-. The most important chem. reactions for the degradation of OMPs are identified for different water matrixes.

Duan, Xiaodi published the artcileDecomposition of Iodinated Pharmaceuticals by UV-254 nm-assisted Advanced Oxidation Processes, Recommanded Product: Diatrizoic Acid, the main research area is decomposition iodinated pharmaceutical UV irradiation advanced oxidation wastewater; Diatrizoate; Hydrogen peroxide; Persulfate; Thyroxine; UV54nm photolysis.

Iodinated pharmaceuticals, thyroxine (a thyroid hormone) and diatrizoate (an iodinated x-ray contrast medium), are among the most prescribed active pharmaceutical ingredients. Both of them have been reported to potentially disrupt thyroid homeostasis even at very low concentrations Here, UV-254 nm-based photolysis and photochem. processes, i.e., UV only, UV/H2O2, and UV/S2O82-, were evaluated for the destruction of these 2 pharmaceuticals. Approx. 40% of 0.5μM thyroxine or diatrizoate was degraded through direct photolysis at UV fluence of 160 mJ/cm2, probably resulting from the photosensitive cleavage of C-I bonds. While the addition of H2O2 only accelerated the degradation efficiency to a low degree, the destruction rates of both chems. were significantly enhanced in the UV/S2O82- system, suggesting the potential vulnerability of the iodinated chems. toward UV/S2O82- treatment. Such efficient destruction also occurred in the presence of radical scavengers when biol. treated wastewater samples were used as reaction matrixes. The effects of initial oxidant concentrations, solution pH, as well as the presence of natural organic matter (humic acid or fulvic acid) and alkalinity were also studied. These results provide insights for the removal of iodinated pharmaceuticals in water and/or wastewater using UV-based photochem. processes.

The development of a modified Kornblum oxidation mediated by 2-picoline N-oxide for the synthesis of belzutifan that circumvented safety and stench problems associated with the original clin. supply route conditions was reported. A robust quench for the crude bromination stream with 1,3-dimethoxybenzene and 2,6-lutidine enabled a single-solvent through-process that avoided the isolation of a mutagenic intermediate and resulted in a significantly lower process mass intensity. A ternary-solvent, constant-volume distillation was then used to crystallize the ketone product directly from the reaction mixture

Zhao, Feng published the artcileEnantioselective Synthesis of α-Aryl-β2-Amino-Esters by Cooperative Isothiourea and Broensted Acid Catalysis, Product Details of C9H10O3, the main research area is aminoarylpropanoate enantioselective preparation; enantioselective aminomethylation pentafluorophenyl arylacetate isothiourea hydrochloride catalyst; mechanism enantioselective aminomethylation pentafluorophenyl arylacetate acylisothiouronium intermediate; aminomethylation; cooperative catalysis; isothiourea; mechanistic studies; α-aryl-β-amino acid.

The synthesis of α-aryl-β2-amino esters such as (S)-Bn2NCH2CHPhCO2Me (Bn = PhCH2) through enantioselective aminomethylation of an arylacetic acid ester in high yields and enantioselectivity via cooperative isothiourea and Broensted acid catalysis is demonstrated. The scope and limitations of this process are explored (25 examples, up to 94% yield and 96:4 er), and the method was applied to the syntheses of (S)-venlafaxine hydrochloride and (S)-nakinadine B. Mechanistic studies are consistent with a C(1)-ammonium enolate pathway being followed rather than an alternative dynamic kinetic resolution process. Control studies indicate that (i) a linear effect between catalyst and product er is observed; (ii) an acyl ammonium ion can be used as a precatalyst; (iii) reversible isothiourea addition to an in situ generated iminium ion leads to an off-cycle intermediate that can be used as a productive precatalyst.
