To a solution of tert-butyl 6-amino-3,4-dihydroisoquinoline-2(1H)-carboxylate (27.1) (2.0 g, 8.05 mmol) in DMF (20 mL) was added to 1-(chloromethyl)-4-methoxybenzene (1.5 g, 9.58 mmol) and K2CO3 (1.11 g, 8.05 mmol). The reaction mixture was stirred at 100 C for 3 h under N2. TLC showed the reaction was complete. The reaction mixture was quenched with the addition of water and extracted with ethyl acetate. The combined organic layers were washed with water and brine, dried over sodium sulfate, and concentrated in vacuo to afford the crude product. The crude product was purified by column chromatography (hexane/ethyl acetate: 8/1) to afford tert-butyl 6-((4-methoxybenzyl)amino)-3,4-dihydroisoquinoline-2(1H)-carboxylate (44.1) as a white solid (1.1 g, 37%).
