A new contrast agent was developed by combining two radiodense elements of gold and iodine within a single PAMAM dendrimer-based nanodevice for potential X-ray computed tomog. (CT) imaging applications. Diatrizoic acid (DTA), an iodinated CT contrast agent, was loaded within the generated dendrimer-entrapped gold nanoparticles (Au DENPs) by three mechanisms (ionic complexation, covalent conjugation and phys. encapsulation) simultaneously. The formed Au DENP-DTA nanocomplexes with about 60 DTA mols. and 100 gold atoms loaded within each dendrimer showed good stability in aqueous solution Measurements of the X-ray absorption coefficient showed that the attenuation effect of Au DENP-DTA is much higher than that of both the com. iodine-based contrast agent at the same iodine concentration and pure Au DENPs at the same gold concentration The incorporation of both AuNPs and iodine-containing small mols. resulted in a significant cooperative enhancing effect in X-ray attenuation. The concept of combining multiple radiodense elements in one NP system may open new avenues for development of a novel class of high-performance CT contrast agents for biomedical imaging applications.
