An efficient Ugi-type three-component reaction (U-3CR) of isocyanides, carboxylic acids and 4-substituted Δ1-piperideines for the synthesis of pipecolic amides such as I is reported. Hydrogenation of 4-substituted pyridines (prepared by alkylation of 4-picoline with alkyl bromides) yielded the 4-substituted piperidines or their hydrochloride salts; oxidation of the piperidines with NCS yielded the Δ1-piperideines which were used without purification The method was used to prepare the anticoagulant argatroban.
