An efficient route has been developed for the facile synthesis of functionalized 2-arylpiperidines I (Ar = 2-bromophenyl, thiophen-3-yl, 2H-1,3-benzodioxol-5-yl, etc.) that may be readily modified using straightforward transformations to create collections of novel substituted 2-arylpiperidines I. The approach features a reaction cascade starting with imine formation from a known 3-((trimethylsilyl)methyl)but-3-en-1-amine and a diverse set of aryl aldehydes ArCHO, followed by an acid-catalyzed aza-Sakurai reaction to assemble a series of 2-aryl-4-exo-methylenepiperidines I in a single step from readily available starting materials. The 2-aryl-4-exo-methylenepiperidines I thus prepared were derivatized via N-methylation, N-acetylation, and N-tosylation.
