With the rapid development and complex challenges of chemical substances, new drug synthesis pathways are usually the most effective.33930-63-1,4-Bromo-2-methylisoquinolin-1(2H)-one,as a common compound, the synthetic route is as follows.

For about 3 min, N2 was bubbled through a mixture of N-benzyl-2-methoxy-5-(tetramethyl-1,3,2-dioxaborolan-2-yl)benzamide (51 mg, 0.14 mmol), 4-bromo-2-methylisoquinolin-1(2H)-one (30 mg, 0.13 mmol), aqueous 1M K3PO4 (0.3 mL) and Pd(dppf)Cl2 (10 mg, 0.013 mmol) in dioxane (1.15 mL) which was then microwaved at 100¡ã C. for 1 h. Work up similar to Example 15 and purification by silica gel chromatography, eluting with 5-50percent EA in hexane over 4 min and continuing 50percent isocratic EA gave the title compound (37 mg, 0.14 mmol) as a tan solid in 71percent yield. 1H NMR (400 MHz, DMSO-d6) delta 3.57 (s, 3H), 3.97 (s, 3H), 4.52 (d, J=6.06 Hz, 2H), 7.21-7.37 (m, 6H), 7.47-7.51 (m, 2H), 7.56 (td, J=5.37, 2.15 Hz, 2H), 7.68-7.73 (m, 1H), 7.79 (d, J=2.27 Hz, 1H), 8.33 (d, J=7.83 Hz, 1H), 8.79 (t, J=6.06 Hz, 1H).
