Herein, a cobalt-catalyzed α-hydroxymethylation of amides RCH2C(O)NR1R2 [R = Ph, 3-methoxyphenyl, thiophen-2-yl, etc.; R1 = Me, Et, Ph; R2 = Me, 2-fluorophenyl, 3-methylphenyl, etc.; R1R2 = -(CH2)5-, -(CH2)2O(CH2)2-] and 1-(2,3-dihydro-1H-indol-1-yl)-2-phenylethan-1-one with methanol under mild conditions was reported. Using CoCl2.6H2O as an inexpensive and efficient catalyst, some important bioactive β-hydroxyamides RCHCH2(OH)C(O)NR1R2 and 3-hydroxy-1-(indolin-1-yl)-2-phenylpropan-1-one were obtained in moderate to excellent yields. The developed method features a wide substrate scope and good functional group tolerance. In addition, anticholinergic tropicamide was easily synthesized in this way.
