3,4-Dihydropyrimidin-2-ones (DHPMs) I [R1 = Ph, 3-HOC6H4, 2-O2NC6H4, etc.; R2 = Me, Et, i-Pr; R3 = H, Me] were synthesized by reaction of aromatic aldehydes, β-ketoester and urea/N-Me urea. These compounds I on oxidation with quinolinium chlorochromate as an oxidizing agent resulted into pyrimidin-2-ones II. The synthesized compounds I and II were screened against three test fungi of wheat crop, i.e., Alternaria triticina, Bipolaris sorokiniana and Ustilago segetum tritici by spore germination method. Compounds II showed excellent antifungal activity than compounds I with least ED50 values 42, 93 and 47 μg/mL. Among the tested compounds, compounds II [R1 = 4-O2NC6H4, R2 = i-Pr, R3 = H; R1 = 2,4-di-MeOC6H3, R2 = Et, R3 = Me] were found to be most active. Structure activity relationship showed that electron withdrawing group enhanced their efficacy due to +I effect and vice versa, whereas dehydrogenation improved their potential due to resonance stabilization of structures involved during electronic interactions between proteins of fungus cells and compounds
