An efficient four-component strategy had been developed for the synthesis of a novel series of (E)-N-benzylidene-3-(benzylthio)-5-(3,5-dimethyl-1H-pyrazol-1-yl)-4H-1,2,4-triazol-4-amines I [R = 4-Me, 2,3-(MeO)2, 3,4,5-F3, etc., R1 = Ph, 4-O2NC6H4, CCH, CH=CH2] in good yields. The method is effective for synthesis of the compounds in a one-pot operation. This method is operationally simple, metal-free, and worked under reflux conditions with diverse substrates. Among all the synthesized compounds, I [R = 4-HO, 2-HO-3-EtO, 2-HO-3-MeO-5-O2N, R1 = Ph; R = 4-MeO, 2,3-(MeO)2, 3,4,5-(MeO)3, R1 = 4-O2NC6H4] showed good results in the in silico studies. Compound I [R = 2-HO-3-MeO-5-O2N, R1 = Ph] showed good results in both DNA binding and mol. docking studies.
