In this report the synthesis, characterization and antimicrobial activity evaluation of a series of triazolo-thiadiazinyl coumarin derivatives I (R = Me, Et, CF3; R1 = H, OMe, OEt, etc.; R2 = H, Cl, Br) and II (R3 = H, Cl, Br; R4 = H, Cl, Br, etc.; R5 = 4-MeOC6H4, isoindoline-1,3-dione). The triazolo thiadiazinyl coumarins were synthesized in a facile tandem solvent free one-pot multi component reaction approach by taking various aliphatic/ aromatic/ heterocyclic carboxylic acids, thiocarbohydrazide and substituted 3-(2-bromoacetyl)coumarins. The newly synthesized mols. were confirmed on the basis of phys., anal. and single crystal X-ray diffraction data. The triazolo thiadiazinyl coumarins exhibited excellent antimicrobial activity against Gram pos., Gram neg. and fungi microbial strains. Among the tested compounds, compounds I (R = Et, R1 = H, R2 = Br), I (R = Et, R1 = R2 = Br), II (R1 = R2 = Cl; R3 = isoindoline-1,3-dione), II (R1 = H; R2 = Br; R3 = isoindoline-1,3-dione) and II (R1 = R2 = Br; R3 = isoindoline-1,3-dione) exhibited potent antimicrobial activity at 25μg/mL concentration against Gram pos. and Gram neg. bacterial strains.
